补肾助孕方对大鼠子宫内膜细胞ER、MMP-9、TIMP-1及卵巢黄体细胞分泌激素的影响

【摘要】:研究目的:观察补肾助孕方对米非司酮干预的大鼠黄体期子宫内膜细胞雌激素受体(ER)、基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶抑制因子-1(TIMP-1)的调控及对大鼠黄体颗粒细胞分泌雌二醇(E2)、孕激素(P)水平的影响。探讨补肾助孕方治疗黄体功能不全性不孕的有效机理。研究方法::1、含药血清制备:选用普通雄兔5只,予补肾助孕方药煎液40g/kg灌胃(相当于成人临床剂量的10倍),2次/d,连续3d。第四天末次灌胃1h(灌胃前禁食不禁水12h),局部麻醉颈动脉采血,室温静置3h,3000rpm离心分离血清15min,收集血清,-20℃保存,56℃水浴灭活补体30min,0.22um微孔滤膜过滤除菌,使用前无血清DMEM稀释至5%、20%浓度的含药血清。2、子宫内膜细胞、卵巢黄体细胞的分离、培养:选择体重50-60g未成年雌性大鼠5只,皮下注射PMSG 50U/只,60h卵泡发育成熟,在皮下注射HCG 35U/只,注射后第七天(HCG注射日为第1天),拉颈断髓法处死,摘取卵巢、子宫,分离和培养子宫内膜细胞和卵巢黄体细胞。表型鉴定后将子宫内膜细胞、卵巢黄体细胞培养于含有100U/ml青霉素、100U/ml链霉素、10%空白血清、DMEM培养液中,置于37℃、5%CO2培养箱内培养,每日观察生长情况,贴壁3-5d传代一次。传代用0.25%胰酶+DMEM培养液。3、含药血清干预:加用药干预48h后,提取各组细胞蛋白、RNA及细胞上清液,观察补肾助孕方含药血清对子宫内膜细胞ERα、ERβ、MMP-9、TIMP-1及卵巢颗粒细胞分泌雌、孕激素水平的影响。Western blot方法检测子宫内膜细胞ER α、ERβ蛋白表达,RT-PCR法检测子宫内膜细胞ERα、ERβ、MMP-9、TIMP-1mRNA的表达量,放射免疫法检测黄体颗粒细胞上清液雌、孕激素的水平。研究结果:(1)补肾助孕方含药血清对子宫内膜细胞ERα、ERβ蛋白的表达有提高作用;同时明显改善米非司酮所致的ERα、ERβ蛋白的低表达;(2)补肾助孕方含药血清对子宫内膜细胞ERα、ERβ、MMP-9、TIMP-1mRNA的表达有提高作用;同时明显改善米非司酮所致的ER、MMP-9、TIMP-1的低表达;(3)补肾助孕方含药血清明显提高卵巢黄体细胞分泌雌、孕激素水平。研究结论:(1)补肾助孕方通过提高子宫内膜细胞ER、MMP-9、TIMP-1的表达,改善子宫内膜的容受性,有利于胚胎着床妊娠,为孕卵着床奠定基础;(2)补肾助孕方药能显著提高卵巢黄体细胞分泌E2、P水平,改善米非司酮所致的E2、P水平低表达,改善米非司酮所致的卵巢黄体功能下降,提高黄体功能,有利于胚胎着床和妊娠维持,提高临床妊娠率。

Effects of estradiol (E 2) secretion and progesterone (P) levels. To explore the effective mechanism of tonifying kidney and helping pregnancy in the treatment of luteal insufficiency infertility. Methods: 5 male rabbits were treated with 40g/kg (equivalent to 10 times of adult clinical dose) twice a day for 3 days. At the end of the fourth day, blood was collected from carotid artery under local anesthesia for 1 hour (12 hours before fasting). Serum was centrifuged at room temperature for 3 h and 3000rpm.
