电针“神门”、“太渊”穴对急性心肌缺血家兔事件相关电位P50、MMN的影响

【摘要】:目的:运用ERPs技术,观察电针手少阴心经原穴“神门”与电针手太阴肺经原穴“太渊”对急性心肌缺血家兔事件相关电位P50和MMN幅值与潜伏期的影响,比较“神门”和“太渊”穴对大脑感觉门控能力(P50)、信息自动加工(MMN)影响的特异性,为明确针刺效应与脑联系的机制提供新的实验依据。 方法:选用成年健康青紫兰家兔50只,雌雄不拘,分笼饲养,体质量(2.5±0.5)kg,,随机选10只作正常对照组,其余家兔在麻醉状态下(8%水合氯醛,1.8ml/kg),采用左冠状动脉前降支结扎法复制急性心肌缺血家兔模型,并将模型复制成功的家兔随机分为模型组、电针“神门”组和电针“太渊”组,每组10只。各电针组刺激电流为1mA,频率为2Hz,每次刺激15min,进行电针治疗。电针结束后,利用美国NEUROSCANERPs工作站,其中E-prime2.0作为刺激器,采用Scan4.3记录并对记录的数据进行处理分析。采用Spass17.0forWindows进行数据统计分析,比较各组家兔大脑皮层听感觉门控电位P50、失匹配负波MMN各指标的变化情况。 结果: (1)Powerlab心电图:与正常组比较,造模后家兔心率显著性下降,ST段显著性抬高,T波倒置。 (2)感觉门控电位P50:各组S1-P50潜伏期和幅值、S2-P50潜伏期比较均无差异(P0.05)。与正常组比较,模型组S2-P50幅值、S2/S1增大(均P<0.01),S1-S2、100*(1-S2/S1)减小(均P<0.01);与模型组比较,电针“神门”组家兔S2-P50幅值、S2/S1减小(均P<0.01),S1-S2、100*(1-S2/S1)增大(均P<0.01),电针“太渊”组无差异(均P0.05)。“神门”组与“太渊”组之间S2-P50幅值、S2/S1、S1-S2、100*(1-S2/S1)差异有统计学意义(P<0.01)。 (3)失匹配负波(MismatchnegativityMMN):与正常组比,模型组潜伏期延长,幅值减小(均P<0.01);与模型组比,“神门”组潜伏期缩短、幅值升高(均P<0.01),“太渊”组比较无统计学意义(均P0.05);“神门”组与“太渊”组之间的差异具有统计学意义(P<0.01)。 结论: (1)急性心肌缺血状态下的家兔大脑感觉门控(P50)、信息自动加工能力(MMN)存在异常变化。 (2)电针不同经穴对急性心肌缺血家兔大脑皮层感觉门控的调整作用有相对特异性,电针心经原穴“神门”的调整作用优于电针肺经原穴“太渊”。 (3)电针不同经穴对急性心肌缺血家兔大脑信息自动加工能力的影响具有特异性,电针心经原穴“神门”的调整作用优于电针肺经原穴“太渊”。
[Abstract]:Objective: to observe the effects of electroacupuncture on the event-related potential P50, MMN amplitude and latency of acute myocardial ischemia in rabbits with acute myocardial ischemia by electroacupuncture at Shenmen, the original point of Shaoyin and Xinjing, and Taiyuan, which is the original point of Taiyin lung meridian. The effects of "Shenmen" and "Taiyuan" on brain sensory gating ability (P50) and automatic processing of (MMN) were compared, which provided a new experimental basis for clarifying the mechanism of acupuncture effect and brain connection. Methods: 50 adult healthy blue rabbits, male and female, were raised in cage, and the body weight was (2.5 卤0.5) kg. 10 rabbits were randomly selected as normal control group. Under anesthesia (8% chloral hydrate 1.8ml / kg), the rabbit model of acute myocardial ischemia was established by ligating the anterior descending branch of left coronary artery. The rabbits were randomly divided into three groups: model group, electroacupuncture "Shenmen" group and electroacupuncture "Taiyuan" group. 10 rats in each group. The stimulation current of each electroacupuncture group was 1 Ma and the frequency was 2 Hz. At the end of electroacupuncture, the American NEUROSCANERPs workstation was used, in which E-prime2.0 was used as stimulator, Scan4.3 was used to record and process the recorded data. To compare the changes of auditory sensory gated potential (P50) and mismatched negative wave (MMN) in the cerebral cortex of rabbits with Spass17.0forWindows. Results: (1) Powerlab electrocardiogram: compared with the normal group, there was no significant difference in the latency of S1-P50 and the amplitude of S2-P50 (P0.05) after the establishment of the model, the significant decrease of heart rate and the elevation of T wave inversion of St segment in the model group. (2) the sensory gated potential P50: there was no difference in the latency of S1-P50 and the amplitude of S2-P50 in each group (P0.05). Compared with the normal group, the amplitude of S2-P50 in the model group increased (P < 0. 01) and the S1-S2100 * (1-S2/S1) decreased (all P < 0. 01), and the amplitude of S2-P50 in the "Shenmen" group decreased (all P < 0. 01), the value of S1-S2100 * (1-S2/S1) increased (all P < 0. 01), but there was no difference in the electroacupuncture "Taiyuan" group (P0.05). There was a significant difference in S2-P50 amplitude between "Shenmen" group and "Taiyuan" group (1-S2/S1) (P < 0. 01). (3). Compared with normal group, the latent period of model group was prolonged and the amplitude decreased (P < 0. 01), while that of model group was higher than that of model group. The latent period of "Shenmen" group was shortened and the amplitude was increased (all P < 0.01), but there was no significant difference between "Taiyuan" group and "Shenmen" group (P0.05), and the difference between "Shenmen" group and "Taiyuan" group was statistically significant (P < 0.01). Conclusion: (1) there are abnormal changes in sensory gating (P50) and information automatic processing ability (MMN) in the brain of rabbits with acute myocardial ischemia. (2) electroacupuncture at different acupoints can affect the sensory function of cerebral cortex in rabbits with acute myocardial ischemia. The regulation of gating has relative specificity. The regulating effect of electroacupuncture on the original acupoint "Shenmen" of the heart meridian was better than that on the "Taiyuan" of the original point of the electroacupuncture of the lung meridian. (3) the effect of different acupoints of electroacupuncture on the ability of automatic processing of cerebral information in rabbits with acute myocardial ischemia was specific. The regulating effect of the original acupoint Shenmen of electroacupuncture is superior to that of the original acupoint Taiyuan of the lung meridian of electroacupuncture.
