肝细胞生长因子在先天性肥厚性幽门狭窄大鼠幽门中的表达

【摘要】:目的研究肝细胞生长因子(HGF)在先天性肥厚性幽门狭窄(CHPS)大鼠幽门的表达情况,探讨HGF在CHPS起病中的作用。方法将受孕后的SD大鼠随机分为模型组与对照组,自孕期第2天始至新生鼠生后21 d,模型组经尾静脉注射给予L-NAME,对照组经相同途径给予同等剂量的生理盐水。新生鼠生后第1、7、14、21天分别称重,生后第21天麻醉处死,留取幽门组织,HE染色后显微镜下测量幽门肌层厚度。RT-PCR半定量幽门组织的HGF mRNA的表达。结果与对照组新生鼠比较,模型组新生鼠体重增长明显缓慢,体重较低,两组间差异有统计学意义(P0.01)。与对照组新生鼠比较,模型组幽门肌明显肥厚,差异有统计学意义(P0.01)。与对照组新生鼠比较,模型组新生鼠HGF mRNA的表达升高,差异有统计学意义(P0.05)。结论幽门组织中HGF的表达增加可能与CHPS的发病有关。
[Abstract]:Objective to investigate the expression of hepatocyte growth factor (HGF) in (CHPS) rats with congenital hypertrophic pyloric stenosis and the role of HGF in the pathogenesis of CHPS. Methods SD rats were randomly divided into model group and control group. From the second day of pregnancy to 21 days after birth, the model group was injected with L-NAME via tail vein, and the control group was given the same dose of normal saline in the same way. The neonatal rats were weighed on the 1st day after birth and were anesthetized and killed on the 21st day after birth. The thickness of the myometrium of the pylorus and the expression of HGF mRNA in the RT-PCR semi-quantitative pyloric tissue were measured under the microscope after HE staining. Results compared with the control group, the body weight of the newborn rats in the model group was significantly slower and lower than that in the control group (P0.01). Compared with the control group, the model group showed significant hypertrophy of pyloric muscle (P0.01). Compared with the control group, the expression of HGF mRNA in the model group was significantly higher than that in the control group (P0.05). Conclusion the increased expression of HGF in pyloric tissues may be related to the pathogenesis of CHPS.
