蒺藜提取物提高大强度耐力运动大鼠的运动能力及其机制

【摘要】:1.目的 研究蒺藜提取物对大强度耐力运动大鼠运动能力的影响及其机制。 2.方法 32只SD大鼠随机分为4组:安静对照组(C,n=8)、蒺藜组(TT,n=8)、运动组(E,n=8)和蒺藜+运动组(TT+E,n=8)。安静对照组和运动组大鼠自由饮用纯水,而蒺藜组和蒺藜+运动组大鼠自由饮用蒺藜提取物配成的蒺藜饮料,记录每日大鼠的饮用量(大鼠摄取的蒺藜提取物在120mg/kg体重左右)。运动大鼠进行大强度的五周递增负荷运动,每周运动6天。5周耐力运动的倒数第二天进行30min的强度为30m/min的跑台运动,断尾取血,检测运动前和运动后30min、2h的血糖水平。5周耐力运动的最后一天,各组大鼠进行一次力竭运动。力竭运动结束后36h,大鼠麻醉后下腔静脉取血,检测血浆睾酮(Testosterone,T)水平、胰岛素样生长因子1(Insulin-like growth factor1,IGF-1)水平、血红细胞数(RBC)、血红蛋白含量(Hb)。大鼠处死后取腓肠肌、比目鱼肌,秤重,用BCA试剂盒检测总蛋白含量;用western blot方法检测雄激素受体(Androgenreceptor,AR)、胰岛素样生长因子1受体(Insulin-like growth factor receptor,IGF-1R)和肌球蛋白重链(Mysion heavy chain,MHC)的蛋白水平。 3.结果 1)蒺藜提取物提高大强度耐力运动大鼠的运动能力 (1)蒺藜提取物减轻大强度耐力运动大鼠体重降低的程度 实验前各组大鼠体重无显著性差异。从第三周开始,运动组、蒺藜+运动组大鼠的体重显著低于安静对照组(p0.05),该作用一直持续至实验结束(p0.01)。与运动组比较,蒺藜+运动组大鼠的体重在第五周时显著增加(p0.05)。此外,蒺藜组大鼠的体重与安静对照组相比,在各周都无显著性差异。 (2)蒺藜提取物提高大强度耐力运动大鼠的一次力竭运动时间 与对照组相比,蒺藜组大鼠的一次力竭运动时间无显著差异。五周耐力运动后,蒺藜+运动组大鼠的一次力竭运动时间比运动组大鼠显著延长(p0.01),约延长了63.2%。此外,运动训练大鼠的一次力竭运动时间比对照组显著延长(p0.05)。 2)蒺藜提取物提高大强度耐力运动大鼠运动能力的机制 (1)蒺藜提取物提高大强度耐力运动大鼠的血浆睾酮水平和腓肠肌AR的蛋白水平 与安静对照组相比,运动组大鼠的血浆睾酮水平显著降低(p0.01);蒺藜+运动组大鼠的睾酮水平与运动组比较显著增加(p0.01),与对照组相比无显著差异,表明蒺藜提取物能完全逆转大强度运动所造成的血睾酮水平降低。此外,与安静组相比较,运动组大鼠腓肠肌的AR蛋白含量无显著性差异。蒺藜+运动组大鼠腓肠肌的AR蛋白表达水平比运动组的显著升高(p0.05)。 (2)蒺藜提取物提高大强度耐力运动大鼠的血浆IGF-1水平和腓肠肌IGF-1R的蛋白水平 与安静对照组相比,运动组大鼠血浆的IGF-1的水平没有显著性差异。蒺藜+运动组大鼠的IGF-1水平与运动组相比较显著增加(p0.05),表明补充蒺藜提取物能提高大强度运动大鼠血浆的IGF-1水平。此外,运动组与安静组相比较,腓肠肌IGF-1R蛋白含量没有显著性差异,蒺藜+运动组与运动组相比较,腓肠肌IGF-1R蛋白表达水平显著升高(p0.05)。 (3)蒺藜提取物增加大强度耐力运动大鼠腓肠肌的总蛋白浓度和MHC的蛋白水平 与对照组相比,运动组大鼠比目鱼肌和腓肠肌的相对重量均显著增加、且腓肠肌的蛋白浓度也显著增加;而蒺藜组大鼠的比目鱼肌和腓肠肌的相对重量、蛋白浓度均无显著差异。与运动组比较,蒺藜+运动组大鼠腓肠肌的蛋白浓度显著增加、但腓肠肌的相对重量以及比目鱼肌的蛋白浓度都没有显著性差异,而比目鱼肌相对重量却比较意外地下降(p0.05)。此外,与安静组相比,运动组的MHC蛋白水平没有显著性差异。与运动组相比,蒺藜+运动组的MHC蛋白水平显著增高(p0.05)。 (4)藜提取物增加大强度耐力运动和不运动大鼠的一次力竭运动后30min的血糖水平 蒺藜提取物能降低运动和不运动大鼠运动前的血糖水平。蒺藜组与安静组比较、蒺藜+运动组与运动组比较,血糖水平在一次力竭运动后的30min显著增加(分别是p0.05,p0.01),但在运动后2h,各组血糖水平无显著差异。 (5)蒺藜提取物对大强度耐力运动和不运动大鼠的Hb、红细胞数无影响 4.结论: 1)蒺藜提取物延长大强度耐力运动大鼠的一次力竭运动时间,提高其运动能力。 2)蒺藜提取物增强大强度耐力运动大鼠运动能力的机制可能与其增加睾酮/AR水平和IGF-1/IGF-1R水平,,从而使腓肠肌的蛋白浓度、MHC蛋白水平增加,以及增加一次力竭运动后30min的血糖水平有关。 3)蒺藜提取物增强大强度耐力运动大鼠运动能力的机制与Hb和RBC数无关。
