膳食晚期糖基化终产物对大鼠的肾脏损伤作用

【摘要】:晚期糖基化终产物(Advanced Glycation End Products,AGEs)是食品加工和贮藏过程中由于发生美拉德反应或脂肪氧化反应而形成的产物。AGEs被认为是一类重要的尿毒症毒素,是形成糖尿病并发症的重要因素。目前AGEs对肾脏的生物学效应研究,主要集中在人工合成AGEs对体外肾脏细胞或生物体的影响,膳食中AGEs对老鼠肾脏功能作用的研究较少。本试验拟选用健康SD大鼠和糖尿病大鼠为动物模型,通过饲喂不同含量AGEs饲料,研究AGEs对肾脏功能的影响,分析AGEs在糖尿病和健康大鼠中各个器官中的分布,探究AGEs对大鼠血脂水平的影响,以更好的评估膳食AGEs的安全性。主要得出以下结论: (1)随着时间的延长,AGEs能够在健康大鼠血清与肾脏中累积,,且血清与肾脏中AGEs含量随着AGEs摄入量的增加而升高。过多的AGEs摄入导致健康大鼠的肾脏功能受损,并导致肾脏氧化应激水平升高。 (2)摄入的AGEs能够在糖尿病大鼠血清和肾脏中累积,高糖引起的AGEs形成增多和肾功能降低引起的AGEs清除率降低是AGEs在糖尿病大鼠体内累积的原因。摄入的AGEs使得糖尿病大鼠的氧化应激水平增强。 (3)肾脏是膳食AGEs累积的重要器官。AGEs在健康大鼠和糖尿病大鼠各器官中含量由高到低为:肾>肝>心脏>肺>脾脏,随着时间的延长,AGEs在各器官中的累积量增加,且随摄入AGEs量的增加而升高。饲喂同一种饲料时,糖尿病大鼠中各器官的荧光性AGEs和CML均高于健康大鼠。 (4) AGEs能引起糖尿病大鼠和健康大鼠的血脂异常。随着AGEs摄入量增加,血清中总胆固醇、甘油三酯及低密度脂蛋白的含量升高。糖尿病大鼠血清中总胆固醇、甘油三酯及低密度脂蛋白的含量均高于正常鼠,而高密度脂蛋白含量随着AGEs摄入量的增加而降低。
[Abstract]:Advanced Glycation End products products (ages) is a product of Maillard reaction or fat oxidation during food processing and storage. Ages is considered to be an important uremic toxin and an important factor in diabetic complications. At present, the biological effects of AGEs on kidney are mainly focused on the effects of synthetic AGEs on renal cells or organisms in vitro, while the effects of AGEs on kidney function in mice are less studied in diet. In this experiment, healthy SD rats and diabetic rats were selected as animal models. The effects of AGEs on renal function were studied by feeding different contents of AGEs diet, and the distribution of AGEs in various organs of diabetic and healthy rats was analyzed. To explore the effect of AGEs on blood lipid level in rats to evaluate the safety of dietary AGEs. The main conclusions are as follows: 1) ages could accumulate in serum and kidney of healthy rats with time, and the content of AGEs in serum and kidney increased with the increase of AGEs intake. Excessive intake of AGEs resulted in impaired renal function and increased renal oxidative stress in healthy rats. (2) AGEs intake could accumulate in serum and kidney of diabetic rats. The increase of AGEs formation induced by high glucose and the decrease of AGEs clearance caused by decreased renal function were the reasons for the accumulation of AGEs in diabetic rats. Intake of AGEs increased oxidative stress levels in diabetic rats. 3) Kidney is an important organ of dietary AGEs accumulation. Ages content in the organs of healthy rats and diabetic rats is from high to low: kidney > liver > heart > lung > spleen. The accumulation of ages in each organ increases with time. And increased with the increase of AGEs intake. The fluorescence AGEs and CML of each organ of diabetic rats were higher than that of healthy rats. AGEs can cause hyperlipidemia in diabetic rats and healthy rats. As AGEs intake increased, serum total cholesterol, triglyceride and low density lipoprotein levels increased. The contents of total cholesterol, triglyceride and low density lipoprotein in serum of diabetic rats were higher than those of normal rats, while the content of high density lipoprotein decreased with the increase of AGEs intake.
