纳米细菌大鼠肾结石模型肾脏结石形成时间的动态研究

【摘要】:目的采用微计算机断层扫描技术(Mirco-CT)和病理组织学检查动态观察纳米细菌诱导的大鼠肾结石模型肾脏结石形成情况。方法 2015年11月—2016年6月,将60只SPF级Wistar大鼠适应性饲养1周后,随机分为对照组和诱石组,各30只。对照组大鼠一次性尾静脉注射0.9%氯化钠溶液1.2 ml,诱石组大鼠一次性尾静脉注射纳米细菌悬液1.2 ml。两组大鼠注射后第1~10周每周处死3只,收集双侧肾脏待测。采用Mirco-CT检查两组大鼠肾脏,统计第1~10周两组大鼠检测到的肾脏高密度影情况。光学显微镜下观察两组大鼠肾脏晶体的形成情况,统计第1~10周两组大鼠肾脏晶体阳性情况。结果 Mirco-CT扫描结果:第1~10周,对照组大鼠肾脏均未见高密度影;第7~10周诱石组大鼠肾脏可见白色颗粒样高密度影。第1~10周,诱石组大鼠肾脏高密度影检出4只(13.3%),对照组未检出;截至第10周末,两组大鼠肾脏高密度影检出率比较,差异无统计学意义(P=0.056)。病理组织学检查结果:第1~10周,对照组大鼠肾脏内无晶体沉积。第4周及第6~10周诱石组大鼠肾脏可见灰白色晶体。第1~10周,诱石组大鼠肾脏晶体阳性11只(36.7%),对照组均为阴性;截至第10周末,诱石组大鼠肾脏晶体阳性率高于对照组(P0.001)。结论纳米细菌在第4周开始诱导大鼠肾脏形成结晶,这个病理过程在第6~10周持续存在,Mirco-CT检查可见第7~10周形成高密度结石样物质,纳米细菌可能通过早期损伤肾脏诱导结晶形成并进一步产生结石。
[Abstract]:Objective to observe the formation of renal calculi in rat renal calculi induced by nanobacteria by microcomputed tomography (Mirco-CT) and histopathological examination. Methods from November 2015 to June 2016, 60 SPF grade Wistar rats were fed adaptively for one week, and were randomly divided into two groups: control group and induced stone group, 30 rats in each group. Control group rats were injected with 0.9% sodium chloride solution 1.2 ml, once a tail vein injection of nano-bacteria suspension 1.2 ml. Three rats were killed 10 weeks after injection, and bilateral kidneys were collected to be tested. Mirco-CT was used to examine the kidney of the two groups. The formation of renal lens in the two groups was observed under optical microscope, and the positive rate of renal lens in the first 10 weeks of the two groups was counted. Results the results of Mirco-CT: at the 1st week, there were no high density shadows in the kidneys of the control group, and white granulocylike high density shadows were observed in the kidneys of the rats in the induced lithotripsy group at the 7th week. At the 1st week, 4 rats (13.3%) were detected by renal hyperdensitometry in the induced lithotripsy group, but not by the control group, but there was no significant difference between the two groups at the end of the 10th week (P0. 056). Results of histopathological examination: at the 1st week, there was no crystal deposition in the kidney of the control group. Grayish white crystals could be seen in the kidney of the rats in the 4 th and 6 th week induced lithotripsy group. At the first week of 10 weeks, 11 rats (36.7%) were positive for renal lens in the induced lithotripsy group, and all the rats in the control group were negative, and at the end of the 10th week, the positive rate of the kidney lens in the induced lithotripsy group was higher than that in the control group (P0.001). Conclusion Nano bacteria began to induce the formation of crystals in the kidney of rats at the 4th week. This pathological process persisted at 6 and 10 weeks, and high density lithium-like substance was found on Mirco-CT examination at 7 ~ 10 weeks. Nano-bacteria may induce crystallization through early kidney injury and further produce stones.
