RNA干扰ROR-γt表达对大鼠肾移植模型中Th17细胞功能和慢性排斥反应的作用

【摘要】:目的探讨RNA干扰Th17关键转录因子ROR-γt表达后,对大鼠肾移植慢性排斥模型中Th17细胞功能以及慢性排斥反应的影响。方法构建ROR-γt特异性干扰腺病毒,将其通过尾静脉注入大鼠慢性排斥反应模型,监测大鼠血肌酐变化,酶联免疫吸附试验检测Th17相关细胞因子IL-17、IL-6和TNF-γ改变。4周后检测大鼠肾组织病理改变。结果成功构建ROR-γt特异性干扰腺病毒pSES-HUS-ROR-γt。大鼠慢性排斥反应模型尾静脉注入pSES-HUS-ROR-γt,大鼠血清肌酐水平下降,IL-17、IL-6和TNF-γ水平下降,肾小球纤维化病变减轻。结论 pSES-HUS-ROR-γt特异性干扰腺病毒可以抑制Th17细胞功能,减轻肾移植慢性排斥反应。
[Abstract]:Objective to investigate the effects of RNA interference on the expression of ROR- 纬 t, a key transcription factor of Th17, on the function of Th17 cells and chronic rejection in chronic renal allograft rejection model in rats. Methods ROR- 纬 t specific interfering adenovirus was constructed and injected into rat model of chronic rejection through tail vein. The changes of serum creatinine in rats were monitored, and the Th17 related cytokine IL-17, was detected by enzyme linked immunosorbent assay (Elisa). The changes of IL-6 and TNF- 纬 were detected after 4 weeks. Results ROR- 纬 t specific interfering adenovirus pSES-HUS-ROR- 纬 t was successfully constructed. PSES-HUS-ROR- 纬 t was injected into the caudal vein of chronic rejection model in rats. Serum creatinine level decreased, IL-17,IL-6 and TNF- 纬 levels decreased and glomerular fibrosis was alleviated. Conclusion pSES-HUS-ROR- 纬 t interference with adenovirus can inhibit the function of Th17 cells and alleviate the chronic rejection of renal transplantation.
