原花青素对碘酸钾致甲状腺氧化损伤的防御作用及机制

【摘要】:目的探讨原花青素对碘酸钾导致小鼠甲状腺氧化损伤的保护作用。方法小鼠按体重随机分为5组,空白对照(NI)组、高碘对照(HI)组、原花青素低剂量(PL)组、原花青素中剂量(PM)组、原花青素高剂量(PH)组。PL、PM、PH组分别给予不同剂量原花青素。6 w后同时摘眼球取血,测血清超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量,并取甲状腺组织做普通病理切片。结果 HI组与NI组比较,呈现明显的抗氧化损伤及甲状腺病理学改变。与HI组比较,PL、PM和PH组中GSH-Px活性代偿性增高的程度稍有下降;SOD活性及SOD/MDA比值均显著增加(P0.05);PM组和PH组中MDA含量显著降低(P0.05)。PM及PH组小鼠的甲状腺切片无显著病理学改变。结论原花青素可显著提高机体的抗氧化能力并保护甲状腺细胞的正常形态,从而使小鼠甲状腺免受高碘导致的氧化损伤。
[Abstract]:Objective to investigate the protective effect of procyanidins on thyroid oxidative damage induced by potassium iodate in mice. Methods mice were randomly divided into 5 groups according to their body weight: blank control group, high iodine control group, low dose proanthocyanidin group, middle dose procyanidin group and PMM group. The proanthocyanidins high dose PH. PLP PMN PH group were given different doses of proanthocyanidins at the same time after 6 weeks of extraction of eyeball blood, serum superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activity and malondialdehyde (MDA) content were measured. Results the HI group was compared with NI group. In comparison with HI group, the degree of compensatory increase of GSH-Px activity was slightly decreased. Sod activity and SOD/MDA ratio were significantly increased in P0.05 group and PH group, and the content of MDA in P0.05 group and PH group was significantly decreased. There were no significant pathological changes in thyroid sections in P0.05, PM and PH groups. Conclusion Proanthocyanidins can significantly improve the antioxidant capacity of the body and protect the normal morphology of thyroid cells. Thus, the thyroid gland of mice is protected from oxidative damage caused by iodine excess.
