运动与白藜芦醇对老年肥胖大鼠视黄醇结合蛋白4、血糖和胰岛素敏感性的影响

【摘要】:目的探讨运动与白藜芦醇对老年肥胖大鼠内脏脂肪组织视黄醇结合蛋白4(RBP4)mRNA和蛋白表达、血浆RBP4浓度、血糖及胰岛素敏感性的影响。方法建立SD老年肥胖大鼠模型,选取自然生长老年大鼠6只为正常组,将24只肥胖鼠随机分为肥胖对照、运动、白藜芦醇、运动+白藜芦醇4组。白藜芦醇为52.5 mg/(kg·d)灌胃,5次/周,持续干预8周。干预后测血糖、胰岛素敏感性、内脏脂肪组织RBP4 mRNA表达(qRT-PCR)、蛋白表达(Western blot)及血浆RBP4浓度(ELISA)。结果 8周干预后,(1)RBP4 mRNA表达:肥胖组(2.63±0.45)高于正常组(2.10±0.15)(P0.05);白藜芦醇组(1.84±0.33)、运动组(1.91±0.15)和运动+白藜芦醇组(2.13±0.11)低于肥胖组(P0.05,P0.01)。(2)RBP4蛋白表达:肥胖组(1.346±0.025)高于正常组(1.196±0.017)(P0.01);运动组(1.025±0.006)低于肥胖组(P0.01)、白藜芦醇组(0.735±0.015)和运动+白藜芦醇组(0.701±0.018)低于运动组(P0.01);运动+白藜芦醇组低于白藜芦醇组(P0.05)。(3)血浆RBP4浓度:肥胖组[(16.00±1.54)μg/L]高于正常组[(13.02±2.20)μg/L](P0.01);运动组[(14.76±1.56)μg/L]低于肥胖组,但差异无统计学意义;白藜芦醇组[(13.59±0.07)μg/L]和运动+白藜芦醇组[(12.98±1.69)μg/L]低于肥胖组(P0.05,P0.01)。(4)血糖:肥胖组[(17.93±6.09)mmol/L]高于正常组[(11.64±3.57)mmol/L](P0.01);运动组[(13.36±1.82)mmol/L]低于肥胖组(P0.05),白藜芦醇组[(15.24±2.19)mmol/L]、运动+白藜芦醇组[(13.95±2.26)mmol/L]低于肥胖组,但差异无统计学意义。(5)胰岛素敏感性:肥胖组(0.37±0.02)低于正常组(0.39±0.02)(P0.05);白藜芦醇组(0.38±0.01)、运动组(0.38±0.02)高于肥胖组,但差异无统计学意义,运动+白藜芦醇组(0.39±0.15)高于肥胖组(P0.05)。上述指标(除蛋白表达外)干预各组间差异无统计学意义。结论老年肥胖大鼠内脏脂肪组织RBP4 mRNA、蛋白表达、血浆RBP4浓度及血糖均增高,胰岛素敏感性降低;运动与白藜芦醇均能降低其RBP4 mRNA、蛋白表达和血浆浓度;单纯运动能明显降低其血糖,运动结合白藜芦醇能明显提高其胰岛素敏感性。在降低蛋白表达、血浆RBP4浓度和提高胰岛素敏感性中,运动结合白藜芦醇表现出协同性。
[Abstract]:Objective to investigate the effects of exercise and resveratrol on the expression of retinol binding protein 4 (RBP4N) mRNA and protein, plasma RBP4 concentration, blood glucose and insulin sensitivity in visceral adipose tissue of aged obese rats. Six aged naturally growing rats were selected as normal group. 24 obese rats were randomly divided into 4 groups: obese control group, exercise group, resveratrol group and exercise group. Resveratrol was given intragastrically for 5 times a week. Continuous intervention for 8 weeks. Blood glucose and insulin sensitivity were measured after intervention. Results the expression of RBP4 mRNA in visceral adipose tissue was significantly higher than that in normal group (2.63 卤0.45), resveratrol group was 1.84 卤0.33, exercise group was 1.91 卤0.15) and exercise group was 2.13 卤0.11) after 8 weeks of intervention, the expression of RBP4 mRNA was significantly lower in obese group than in normal group (2.63 卤0.45), while in resveratrol group (1.84 卤0.33), exercise group (1.91 卤0.15) and exercise group (2.13 卤0.11) were significantly lower than those in normal group. The expression of RBP4 protein in obese group (1.346 卤0.025) was higher than that in normal group (1.196 卤0.017), in exercise group (1.025 卤0.006) was lower than that in obese group (0.735 卤0.015) and in exercise group (0.701 卤0.018) was lower than that in exercise group. Degree: the obese group [16.00 卤1.54 渭 g / L] was higher than the normal group [13.02 卤2.20 渭 g / L] P0.01g / L; the exercise group [14.76 卤1.56 渭 g / L] was lower than the obese group. But there was no significant difference in blood glucose: the blood glucose levels in the resveratrol group [13.59 卤0.07 渭 g / L] and the exercise resveratrol group [12.98 卤1.69 渭 g / L] were lower than those in the obese group (P 0.05 P 0.01g / L): the obese group [17.93 卤6.09 mmol / L] was higher than the normal group [11.64 卤3.57 mmol / L] P0.01 / L; the exercise group [13.36 卤1.82 mmol / L] was lower than the obese group; the resveratrol group [15.24 卤2.19 mmol / L], the exercise group [13.95 卤2.26 mmol / L] were lower than the obese group. But the insulin sensitivity of obese group was 0.37 卤0.02) lower than that of normal group 0.39 卤0.02P0.05, resveratrol group 0.38 卤0.01g, exercise group 0.38 卤0.02), but the difference was not statistically significant. Exercise resveratrol group (0.39 卤0.15) was higher than obese group (P 0.05). There was no significant difference between the above indexes (except protein expression). Conclusion the expression of RBP4 mRNAs, protein expression, plasma RBP4 concentration and blood sugar in visceral adipose tissue of aged obese rats were increased. Both exercise and resveratrol could decrease RBP4 mRNAs, protein expression and plasma concentration, exercise alone could significantly decrease blood glucose, exercise combined with resveratrol could significantly increase insulin sensitivity. Exercise combined with resveratrol showed synergism in plasma RBP4 concentration and insulin sensitivity.
