2型糖尿病小鼠周围神经病变模型的建立及皮下神经纤维密度的检测

【摘要】:目的建立2型糖尿病小鼠周围神经病变模型,并对该模型的皮下神经纤维密度(intra-epidermal nerve fiber density,IENFD)进行检测。方法 C57BL6雄性小鼠24只,随机分为四组,每组6只:HS组采用高脂饲料喂养24周+链脲佐菌素STZ(120 mg/kg)单次注射;H组采用高脂饲料喂养24周+缓冲液注射;S组普通饲料喂养24周+STZ(120 mg/kg)单次注射;C组普通饲料喂养24周+缓冲液注射。四组小鼠均以24周为实验终点,测定随机血糖、胰岛素抵抗指数(HOMA-IR)、机械痛阈和IENFD。结果第24周时HS组随机血糖、HOMA-IR明显高于H、S、C组(P0.01),机械痛阈明显低于H、S、C组(P0.05),IENFD明显低于S组和C组(P0.05);H组随机血糖、HOMA-IR明显高于C组(P0.01),机械痛阈与S组、C组差异无统计学意义,IENFD与HS组差异无统计学意义。结论高脂饲料喂养联合中等剂量STZ成功建立了2型糖尿病小鼠周围神经病变模型,小鼠皮下神经纤维密度明显下降。
[Abstract]:Objective to establish a model of peripheral neuropathy in type 2 diabetic mice and to detect the subcutaneous nerve fiber density of intra-epidermal nerve fiber density.Methods Twenty-four male C57BL6 mice were randomly divided into four groups. Each group of 6 HS rats were fed high fat diet for 24 weeks with streptozotocin STZ(120 mg / kg) single injection of streptozotocin (STZ(120 mg / kg). Group H was fed with high fat feed for 24 weeks. Group S was fed with normal diet for 24 weeks. Group C was fed with normal diet for 24 weeks. After 24 weeks of culture, the mice were injected with buffer solution for 24 weeks. Random blood glucose was measured, Results at the 24th week, the HOMA-IR of HS group was significantly higher than that of P0.01C group, and the mechanical pain threshold was significantly lower than that of P0.05- IENFD in group C and P0.05H group. The HOMA-IR in HS group was significantly higher than that in group C, and the mechanical pain threshold in group C was significantly higher than that in group C (P0.05H group), and the level of HOMA-IR in group C was significantly higher than that in group C (P 0.01), and the mechanical pain threshold in group C was significantly lower than that in group C and group S (P 0.05H). There was no significant difference between group C and group S in pain threshold. There was no significant difference between IENFD and HS group. Conclusion the model of peripheral neuropathy in type 2 diabetic mice was successfully established by high fat diet and moderate dose STZ. The density of subcutaneous nerve fibers decreased significantly in mice.
