骨髓间充质干细胞对苯所致小鼠造血损伤修复作用

【摘要】:目的探讨骨髓间充质干细胞(BMSCs)对苯中毒小鼠造血损伤的修复作用。方法取无特定病原体级健康雄性昆明小鼠5只,以全骨髓贴壁法分离培养BMSCs。取同类小鼠,以苯油混合物进行多点皮下注射染毒(连续10周,每周3次),建立苯中毒动物模型后,随机分为苯中毒组和BMSCs移植组,每组18只,分别经尾静脉输注250.0μL的0.9%氯化钠溶液或细胞密度为2×10~9/L的BMSCs,每周1次,连续治疗4周。另设对照组小鼠(10只),不予任何处理。治疗结束2周后处死各组小鼠,检测血常规,以瑞氏-吉姆萨法染色观察小鼠骨髓有核细胞,计数骨髓造血祖细胞集落数量,以酶联免疫吸附实验检测小鼠血清中γ-干扰素(IFN-γ)水平。结果成功制备苯中毒动物模型。BMSCs移植疗程结束后,苯中毒组小鼠周围血中白细胞计数、血小板计数、红细胞计数、血红蛋白水平和骨髓中有核细胞总数、粒细胞-巨噬细胞集落形成单位(CFU-GM)均低于对照组(P0.01),BMSCs治疗组小鼠上述指标均高于苯中毒组(P0.05);BMSCs治疗组小鼠血小板计数、红细胞计数和骨髓中有核细胞总数、CFU-GM均低于对照组(P0.05)。苯中毒组小鼠血清中IFN-γ水平高于对照组(P0.01);BMSCs治疗组小鼠该指标低于苯中毒组(P0.01),但与对照组比较,差异无统计学意义(P0.05)。结论 BMSCs对苯所致的造血系统损伤具有修复作用。
[Abstract]:Objective to investigate the repair effect of bone marrow mesenchymal stem cells (BMSCs) on hematopoietic injury in benzene intoxicated mice. Methods five healthy male Kunming mice without specific pathogens were isolated and cultured by whole bone marrow adherent method. The animal model of benzene poisoning was established by subcutaneous injection of benzene oil mixture (10 consecutive weeks, 3 times a week). The animal model was randomly divided into benzene poisoning group and BMSCs transplantation group, 18 rats in each group. 0.9% sodium chloride solution of 250.0 渭 L or BMSCs with cell density of 2 脳 10 ~ (9) / L were injected via tail vein once a week for 4 weeks. 10 mice in the control group were treated without any treatment. After 2 weeks of treatment, the mice in each group were sacrificed and their blood routine was detected. The nucleated bone marrow cells of mice were observed by Rish-Jim 's method and the number of hematopoietic progenitor cells in bone marrow was counted. The level of IFN- 纬 in serum of mice was detected by enzyme-linked immunosorbent assay (Elisa). Results the animal model of benzene poisoning was established successfully. After the course of transplantation of BMSCs, the peripheral white blood cell count, platelet count and erythrocyte count in benzene poisoning group were measured. The levels of hemoglobin, the total number of nucleated cells in bone marrow, and the CFU-GM) of granulocyte-macrophage colony forming unit (CFU-GM) were all lower than those of the control group (P 0.01) and the above indexes were higher than those of the group treated with benzene poisoning (P 0.05) and the platelet count of the group treated with BMSCs were higher than those of the group treated with benzene poisoning. The count of erythrocyte and the total number of nucleated cells in bone marrow were lower than that of control group (P 0.05). The level of IFN- 纬 in serum of mice in benzene poisoning group was higher than that in control group (P 0.01), but the level of IFN- 纬 in BMSCs group was lower than that in control group (P 0.01), but it was higher than that in control group. Conclusion BMSCs can repair the damage of hematopoietic system induced by benzene.
