【摘要】:目的探讨中国汉族人群糖皮质激素诱导转录因子1(GLCCI1)基因rs37973位点多态性与哮喘的相关性。方法应用聚合酶链反应(PCR)、DNA测序、哮喘控制测试评分(ACT)及肺功能测定的方法。结果不同种族人群GLCCI1基因rs37973位点等位基因频率的比较差异有统计学意义(X~2=89.07,P0.05)。我国汉族哮喘人群及健康者rs37973位点均存在基因多态现象(表型包括AA、AG和GG三种基因型),各基因型分布频率哮喘组与健康对照组比较差异有统计学意义(X~2=8.27,P0.05);A/G等位基因频率比较差异有统计学意义(X~2=4.14,P0.05)。相对AA基因型而言,AG杂合型、GG+AG均能显著增加哮喘发生的危险性。rs37973位点等位基因频率与哮喘关系表明,相对A而言,突变G能显著增加哮喘发生的危险性。在哮喘组3种基因型的ACT评分、第1秒用力呼气容积(FEV_1)、第1秒用力呼气容积占预计值的百分比(FEV_1/Pred%)、第1秒用力呼气容积占用力肺活量的百分比(FEV_1/FVC%)比较差异无统计学意义(P均0.05)。结论我国汉族人群存在着GLCCI1基因rs37973位点多态性,且其多态性与哮喘患病相关。
[Abstract]:Objective to investigate the association of rs37973 polymorphism of glucocorticoid inducible transcription factor 1 (GLCCI1) gene with asthma in Chinese Han population. Results there were significant differences in allele frequencies of GLCCI1 rs37973 locus in different ethnic groups. There was a significant difference in the allele frequency of rs37973 locus of GLCCI1 gene. The rs37973 loci were found in both Chinese Han asthmatic population and healthy subjects. Because of polymorphism (phenotypes including AAAAG and GG genotypes), the frequency of genotype distribution in asthma group was significantly different from that in healthy control group. There was a significant difference in allele frequency of A- / G allele between the asthma group and the healthy control group. There was a statistically significant difference in the allele frequency of the A / G allele between the AA genotype and the AA genotype. The allelic frequency of allelic locus. Rs37973 and asthma were significantly increased by AG heterozygous GG AG, indicating that GG AG could significantly increase the risk of asthma.
