在有氧运动的基础上服用补肾壮骨颗粒对改善去势大鼠骨质疏松症病情的协同效果

【摘要】:目的探讨有氧运动与补肾壮骨颗粒综合疗法治疗绝经后骨质疏松症(postmenopausal osteoporosis,PMOP)的疗效及机理,为有效防治PMOP提供理论依据。方法采用去势大鼠模型,分为假手术组(n=10)、模型组(n=10)、阳性组(n=10)、有氧运动组(n=10)、补肾壮骨颗粒组(n=10)和联合组(n=10),去势后2 w运用有氧运动、补肾壮骨颗粒以及二者联合等干预方法防治大鼠发生的骨质疏松。连续12 w检测各组大鼠骨组织生物力学,同时采用实时荧光定量PCR技术检测骨代谢相关指标表达。结果与假手术组相比,模型组大鼠的左股骨最大负荷、左胫骨最大负荷和第四腰椎结构强度均显著降低(P0.05)。与模型组相比,阳性组、有氧运动组、补肾壮骨颗粒组和联合组的左股骨最大负荷、左胫骨最大负荷和第四腰椎结构强度均显著提高(P0.05)。荧光定量PCR结果显示,与模型组相比,阳性组、有氧运动组、补肾壮骨颗粒组和联合组的RANKL和OPG均显著提高(P0.05),而RANKL/OPG显著降低(P0.05)。与阳性组相比,联合干预组的RANKL和OPG均显著提高(P0.05),而RANKL/OPG显著降低(P0.05)。结论补肾壮骨颗粒和运动的综合性治疗策略能通过降低骨组织中OPG/RANKL比值,进而抑制破骨细胞的活性来改善去势大鼠骨质疏松症病情,其疗效要优于单纯运动及补肾壮骨颗粒。
[Abstract]:Objective to explore the therapeutic effect and mechanism of aerobic exercise and Bushen Zhuanggu granule in treating postmenopausal osteoporosis (PMOP), and to provide theoretical basis for effective prevention and treatment of PMOP. Methods Ovariectomized rat model was used. They were divided into sham operation group, model group, positive group, aerobic exercise group, Bushen Zhuanggu granule group and combined group. Aerobic exercise was used 2 weeks after castration. Bushen Zhuanggu granule and its combination were used to prevent and treat osteoporosis in rats. Biomechanics of bone tissue in each group was detected for 12 weeks. At the same time, real-time fluorescence quantitative PCR was used to detect the expression of bone metabolism related indexes. Results compared with sham operation group, the maximal load of left femur in model group was higher than that in sham operation group. The maximum load of left tibia and the strength of the fourth lumbar vertebrae decreased significantly compared with the model group, the maximal load of left femur in positive group, aerobic exercise group, Bushen Zhuanggu granule group and combined group. The maximum load of left tibia and the structural strength of the fourth lumbar vertebra increased significantly. The results of fluorescence quantitative PCR showed that, compared with the model group, positive group, aerobic exercise group, Both RANKL and OPG in Bushen Zhuanggu granule group and combined group were significantly increased, while RANKL/OPG significantly decreased P0.05. In the combined intervention group, both RANKL and OPG increased significantly, while RANKL/OPG significantly decreased P0.05.Conclusion the comprehensive treatment strategy of Bushen Zhuanggu granule and exercise can reduce the ratio of OPG/RANKL in bone tissue. And then inhibit the activity of osteoclasts to improve the condition of osteoporosis in ovariectomized rats, its curative effect is better than simple exercise and Bushen Zhuanggu granule.
