多囊卵巢综合征患者颗粒细胞microRNA-200b的表达及影响

【摘要】:目的检测多囊卵巢综合征(PCOS)患者卵巢颗粒细胞中microRNA-200b(miR-200b)的表达,分析其与胰岛素抵抗的关系。方法选取PCOS患者120例(其中胰岛素抵抗78例,非胰岛素抵抗42例)及对照患者72例。采用逆转录定量PCR(qRT-PCR)检测在卵巢颗粒细胞中miR-200b的表达,并分析其临床意义。结果 PCOS胰岛素抵抗患者中超重或肥胖、高血压、高血糖、脂代谢紊乱和代谢综合征的发生率皆高于PCOS非胰岛素抵抗患者和对照患者(P均0.05)。PCOS胰岛素抵抗患者miR-200b表达上调,与PCOS非胰岛素抵抗患者和对照患者比较差异有统计学意义(P=0.012,P0.001);且PCOS胰岛素抵抗患者中miR-200b表达量与胰岛素抵抗指数(HOMA-IR)呈正相关(r=0.532,P=0.002)。结论 PCOS胰岛素抵抗患者高血压等代谢方面远期并发症的患病率较高。miR-200b高表达与PCOS胰岛素抵抗呈正相关,可能是PCOS患者出现胰岛素抵抗的重要病理机制之一。
[Abstract]:Objective to detect the expression of microRNA-200bmmiR-200b in granulosa cells of patients with polycystic ovary syndrome (PCOS) and to analyze the relationship between microRNA-200bmmiR-200b and insulin resistance.Methods 120 patients with PCOS (78 with insulin resistance and 42 with non-insulin resistance) and 72 controls were selected.The expression of miR-200b in ovarian granulosa cells was detected by reverse transcription quantitative PCR qRT-PCR and its clinical significance was analyzed.Results the incidences of overweight or obesity, hypertension, hyperglycemia, lipid metabolism disorder and metabolic syndrome in patients with PCOS insulin resistance were higher than those in patients with PCOS and control group. MiR-200b expression was up-regulated in patients with 0.05).PCOS insulin resistance.There was a significant difference between the PCOS patients with non-insulin resistance and the controls, and there was a positive correlation between the expression of miR-200b and the insulin resistance index (HOMA-IRI) in the patients with PCOS insulin resistance, and there was a positive correlation between the expression of miR-200b and the insulin resistance index (HOMA-IRI) in the patients with PCOS insulin resistance.Conclusion the high expression of .miR-200b in patients with PCOS insulin resistance is positively correlated with PCOS insulin resistance, which may be one of the important pathological mechanisms of insulin resistance in PCOS patients.
