下丘脑室旁核orexin-A对大鼠体重和自由活动的影响及机制

【摘要】:目的:探讨室旁核(PVN)微量注射orexin-A对大鼠体重的影响及机制。方法:采用PVN埋管、PVN微量注射,Echo MRI900体成分分析仪分析身体成分等方法,观察室旁核微量注射orexin-A对大鼠体重、身体成分和自由活动的影响。结果:与注射人工脑脊液对照组相比,下丘脑PVN微量注射orexin-A,大鼠体重明显减轻(P0.05),但大鼠摄食量无明显差异(P㧐0.05)。实验结束后6天,PVN微量注射orexin-A组大鼠体重仍显著低于注射前(P0.05),而人工脑脊液对照组大鼠体重比注射前显著增加(P0.05);与注射人工脑脊液对照组大鼠比较,PVN微量注射Orexin-A,大鼠肌肉组织、脂肪组织和非脂肪组织均显著降低(P0.05),但注射orexin-A的大鼠降低更明显。与注射人工脑脊液对照组相比,PVN微量注射orexin-A,大鼠的摄食转化率(摄食量/体重)显著降低(P0.05)。下丘脑PVN微量注射orexin-A,大鼠水平运动量显著增加(P0.05)。结论:室旁核orexin-A可通过增加大鼠活动量而产生负能量平衡,体重减轻。
[Abstract]:Aim: to investigate the effect and mechanism of microinjection of PVN into paraventricular nucleus (PVN) on body weight of rats.Methods: the effects of microinjection of orexin-A into paraventricular nucleus on body weight, body composition and free activity of rats were observed by means of microinjection of Echo MRI900 body composition analyzer.Results: compared with the control group injected with artificial cerebrospinal fluid (ACSF), hypothalamic PVN microinjection of orexin-A significantly reduced the body weight of the rats, but there was no significant difference in the feeding intake of the rats.Six days after the experiment, the body weight of rats in the orexin-A group was still significantly lower than that in the pre-injection group, while the body weight of the rats in the artificial cerebrospinal fluid control group was significantly higher than that in the control group before injection, and compared with the control group of artificial cerebrospinal fluid injection of Orexin-A, the muscle tissue of the rats was significantly higher than that of the control group.Both adipose tissue and non-adipose tissue significantly decreased P0. 05%, but more significantly in rats injected with orexin-A.Compared with the control group injected with artificial cerebrospinal fluid (ACSF), the feed conversion rate (feed intake / body weight) of PVN was significantly lower than that of the control group treated with microinjection of orexin-A.After microinjection of orexin-A into hypothalamus PVN, the amount of horizontal exercise in rats increased significantly (P 0.05).Conclusion: paraventricular nucleus orexin-A can produce negative energy balance and weight loss by increasing the activity of rats.
