儿茶素和左甲状腺素钠对甲状腺功能减退大鼠氧化应激和认知能力的影响

【摘要】:目的观察左甲状腺素钠、儿茶素以及两药联合治疗对甲状腺功能减退模型大鼠认知能力的影响,对血清8-羟基脱氧鸟苷(8-OHd G)、对氧磷酶-1含量的影响以及海马组织凋亡蛋白caspase-3含量的影响。方法选取50只成年SD雄性大鼠,随机选取10只作为对照组,剩余40只用丙基硫氧密啶(PTU)诱导甲状腺功能减退大鼠模型。PTU组、儿茶素组、左甲状腺素钠组(L-T4组)、左甲状腺素钠+儿茶素组(联合用药组)。采用Morris水迷宫系统进行大鼠行为学检测。采用酶联免疫吸附试验测定各组大鼠血清8-OHd G、对氧磷酶-1水平,采用Western blot测定各组大鼠海马组织凋亡蛋白caspase-3的含量。结果一、行为学检测与对照组比较,PTU组大鼠逃避潜伏期明显延长,差异有统计学意义(P0.05);与PTU组比较,L-T4组、联合用药组大鼠逃避潜伏期均明显缩短,差异有统计学意义(P0.05)。二、指标检测(1)与对照组比较,PTU组大鼠血清8-OHd G含量明显升高,差异有统计学意义(P0.05);与PTU组比较,L-T4组、联合用药组大鼠血清8-OHd G含量明显降低,差异有统计学意义(P0.05)。?与对照组比较,PTU组大鼠血清对氧磷酶-1含量明显降低,差异有统计学意义(P0.05);与PTU组比较,L-T4组、联合用药组大鼠血清对氧磷酶-1含量明显升高,差异有统计学意义(P0.05)。?与对照组比较,PTU组大鼠海马组织凋亡蛋白caspase-3含量明显升高,差异有统计学意义(P0.05);与PTU组比较,LT4组、联合用药组大鼠海马组织凋亡蛋白caspase-3含量明显降低,差异有统计学意义(P0.05)。结论甲状腺功能减退可降低机体的抗氧化应激能力以及增加细胞的凋亡,左甲状腺素钠、儿茶素药物治疗可以改善氧化应激、减少细胞的凋亡,改善大鼠的认知能力,两药联合使用具有协同的作用。
[Abstract]:Objective to observe the effects of levothyroxine sodium, catechin and combined therapy on the cognitive ability of hypothyroidism rats. The changes of 8-hydroxydeoxyguanosine guanosine 8-OHd (8-OHd), phosphatase-1 and apoptotic protein caspase-3 in hippocampal tissue were studied. Methods 50 adult Sprague-Dawley male rats were selected and 10 rats were randomly selected as control group. The remaining 40 rats were used to induce hypothyroidism model. PTU group and catechin group. The levothyroxine sodium group was treated with L-T4 and the levothyroxine sodium catechin group. The behavior of rats was detected by Morris water maze system. The levels of serum 8-OHd G and paraoxonase-1 were measured by enzyme-linked immunosorbent assay (Elisa), and the content of apoptotic protein caspase-3 (caspase-3) in hippocampal tissue of rats in each group was measured by Western blot. The results were as follows: (1) compared with the control group, the latency of escape in the PTU group was significantly longer than that in the control group, the difference was statistically significant (P 0.05), and that in the L-T4 group was significantly shorter than that in the PTU group (P 0.05). (2) compared with the control group, the serum 8-OHd G level in the control group was significantly higher than that in the control group (P 0.05), and that in the L-T4 group was significantly lower than that in the L-T4 group, and the difference was statistically significant (P 0.05). Compared with the control group, the level of serum paraoxonase-1 in the PTU group was significantly lower than that in the control group, the difference was statistically significant (P 0. 05), and that in the L-T4 group was significantly higher than that in the PTU group, and the difference was statistically significant (P 0. 05, P 0. 05, P 0. 05, P < 0. 05, P < 0. 05, P < 0. 05, P < 0. 05). Compared with the control group, the content of apoptotic protein caspase-3 in hippocampal tissue in PTU group was significantly higher than that in control group (P 0.05), and that in LT4 group was significantly lower than that in LT4 group (P 0.05). Conclusion hypothyroidism can decrease the ability of anti-oxidative stress and increase the apoptosis of cells. The treatment of levothyroxine sodium and catechin can improve the oxidative stress, decrease the apoptosis of cells, and improve the cognitive ability of rats. The combined use of the two drugs has synergistic effect.
