钙对生长期染铅大鼠免疫损伤干预机制的研究

【摘要】:[目的]通过观察不同剂量钙对染铅大鼠体内二价金属元素、白细胞数、血清IgG、脾脏指数和脾脏组织形态,及免疫相关细胞因子基因的mRNA表达量等的影响,探索钙对生长期染铅大鼠免疫损伤的干预机制,进而为有效预防铅中毒提供理论依据。[方法]选用21日龄清洁级SD雄性大鼠60只,适应性喂养7d后,按体重随机分为5组,每组12只,分别为对照组(基础饲料,去离子水)、染铅组(基础饲料,0.2%醋酸铅水)、试验Ⅰ组(基础饲料添加0.5%碳酸钙,0.2%醋酸铅水)、试验Ⅱ组(基础饲料添加1.0%碳酸钙,0.2%醋酸铅水)、试验Ⅲ组(基础饲料添加2.0%碳酸钙,0.2%醋酸铅水)。饲养60d后,采集全血、肝脏、肾脏、脾脏、心脏、肌肉,测定其Pb、Ca、Zn、Fe、Mn、Mg、Cu的含量;制备血清,测定其白细胞数及血清IgG含量;脾脏称重,计算脾脏指数;HE染色观察大鼠脾组织细胞形态学变化;采用Trizol提取脾脏中总的RNA,应用荧光定量RT-PCR法测定脾脏中TNF-α、IL-1β和IL-6基因的mRNA相对表达量。[结果](1)与对照组相比,染铅组大鼠心、肝、脾、肾、肌肉及血液中铅的含量明显升高(P0.05或P0.01);各组织中钙含量均降低,差异显著(P0.05);铁、铜、锌、锰含量在各组织中均降低,脾中铁、肝肾中铜、血中锌含量差异均达显著或极显著(P0.05或P0.01);心脏中镁含量升高,差异极显著(P0.01)。与染铅组比较,试验组大鼠铅含量降低,试验Ⅱ组各组织中降低明显,差异显著或极显著(P0.05或P0.01);各组织中钙含量升高,试验Ⅰ、Ⅱ组达差异显著(P0.05);铜、锌含量在各组织中均有所升高,心、肝、肾、肌肉中的试验Ⅱ组差异显著(P0.05)。表明适量的补钙可以干预铅致动物机体组织中二价矿物元素的失衡。(2)与对照组比较,染铅组大鼠脾脏指数、白细胞总数、淋巴细胞百分比、血清IgG含量下降(P0.05或P0.01),单核细胞百分比和嗜中细胞百分比升高(P0.05或P0.01)。与染铅组比较,试验Ⅱ组的脾脏指数、白细胞数及试验Ⅲ组的血清IgG含量上升,差异均达显著水平(P0.05);单核细胞百分比和嗜中细胞百分比下降,但差异不显著。说明补充适量的钙可以通过调节脾脏指数、白细胞数和血清IgG干预铅对免疫细胞的损伤。(3)与对照组比较,染铅组大鼠脾脏淋巴细胞减少,红白髓交界不明显,有部分发生溶解现象,且淋巴细胞排列稀疏,淋巴小结数量较少。与染铅组比较,试验Ⅱ组大鼠脾脏淋巴细胞增加,红白髓分界比较明显,淋巴细胞基本恢复正常,而试验Ⅲ组淋巴结有明显分散现象。表明补钙可以有效干预染铅导致脾脏病理性变化,但应控制在一定范围内。(4)与对照组比较,染铅组大鼠脾脏中的IL-1β、IL-6、TNF-α的相对表达量降低,差异显著或极显著(P0.05或P0.01);与染铅组比较,三个试验组IL-1β、IL-6、TNF-α的相对表达量均上升,试验Ⅱ组差异极显著(P0.01)。[结论]膳食中补充适量的钙可干预机体对铅的吸收,调节铅致大鼠体内Ca、Zn、 Fe、Mn、Mg、Cu的代谢失衡,调控染铅机体免疫细胞增值能力及脾脏中的IL-1β、IL-6、 TNF-α基因的mRNA相对表达量。可见,补充一定剂量的钙对铅致机体免疫损伤有一定的缓解作用,提高染铅机体免疫能力。
[Abstract]:Objective : To explore the effect of different doses of calcium on the levels of divalent metal elements , white blood cells , serum IgG , spleen index and spleen tissue morphology , and mRNA expression level of immune - related cytokine genes in lead rats . After 60 days of feeding , whole blood , liver , kidney , spleen , heart and muscle were collected , and the contents of Pb , Ca , Zn , Fe , Mn , Mg and Cu were measured .

The mRNA relative expression of TNF - 伪 , IL - 1尾 and IL - 6 in spleen was determined by fluorescence quantitative RT - PCR . The content of lead in heart , liver , spleen , kidney , muscle and blood increased significantly ( P0.05 or P0.01 ) .
The contents of iron , copper , zinc and manganese decreased in all tissues , and the difference of Zn in spleen , iron , liver and kidney was significant or very significant ( P0.05 or P0.01 ) . Compared with the lead - stained group , the lead content in the experimental group was decreased , and the difference was significant or very significant ( P0.05 or P0.01 ) .
