双峰驼奶对2型糖尿病大鼠肝脏和肾脏的组织形态及其细胞凋亡影响的研究

【摘要】:本论文研究了双峰驼奶对2型糖尿病大鼠肝脏和肾脏组织的保护作用,旨在进一步为利用驼奶有效成分在临床上预防和治疗糖尿病及其并发症提供理论参考。通过链脲佐菌素(STZ)结合高糖高脂饮食诱导构建了2型糖尿病(T2DM)大鼠模型,糖尿病模型大鼠随机分为糖尿病模型组(DMC)、低剂量驼奶组(LCM)、高剂量驼奶组(HCM)和盐酸二甲双胍组(MTH),另设正常对照组(C)。试验期4周内,检测各组大鼠的生理生化指标,并采用H.E染色法、免疫组织化学法、免疫印迹法和实时荧光定量法,探讨不同剂量驼奶对大鼠肝脏和肾脏的组织形态和细胞凋亡的影响。结果显示,各实验组糖尿病模型大鼠的体重均呈下降趋势,与DMC组和MTH组相比,LCM组大鼠体重显著降低(P0.01);给予葡萄糖后的120 min内,各组大鼠的血糖浓度均先升高后降低,4个时间点的LCM组和HCM组血糖浓度显著低于DMC组(P0.01),同时,驼奶降低了糖尿病模型大鼠的空腹血糖水平(FPG)和空腹胰岛素(FINS)水平,提高胰岛素敏感指数(ISI);经过驼奶治疗后,肝脏组织部分细胞恢复正常结构,脂肪变性程度明显减轻,没有空泡化现象,肾脏组织基本形态结构完整清晰,肾小球毛细血管无明显扩张,囊腔间隙正常,大部分肾小管上皮细胞形态结构正常;免疫组化和免疫印迹法表明驼奶可抑制Bax和NF-?B蛋白的表达,上调Bcl-2蛋白的表达量;与MTH相比,驼奶可抑制肝脏组织中Bax和NF-?B m RNA的表达量,上调了caspase-3 m RNA的表达量,下调Bcl-2 m RNA的表达量,此外,驼奶还降低肾脏组织中Bax和NF-?B m RNA的表达量,上调Bcl-2 m RNA的表达量,而caspase-3 m RNA在LCM和HCM组的高表达水平,仍待进一步研究。综上所述,驼奶可改善2型糖尿病大鼠的生理生化指标,抑制肝、肾组织相关凋亡蛋白和基因的表达量,结果提示双峰驼奶对2型糖尿病大鼠的肝脏和肾脏组织有一定的保护作用。
[Abstract]:In this paper, the protective effect of bactrian camel milk on liver and kidney tissue of type 2 diabetic rats was studied. The aim of this study was to provide a theoretical reference for the clinical prevention and treatment of diabetes mellitus and its complications by using the effective components of camel milk. A rat model of type 2 diabetes mellitus (T2DM) was established by streptozotocin (STZ) combined with high glucose and high fat diet. Diabetic rats were randomly divided into diabetic model group, low dose camel milk group, high dose camel milk group (HCM) and metformin hydrochloride group, and normal control group. The physiological and biochemical indexes of rats in each group were detected during the four weeks of experiment, and were detected by e staining, immunohistochemistry, immunoblotting and real-time fluorescence quantitative analysis. To investigate the effects of different doses of camel milk on the histomorphology and apoptosis of rat liver and kidney. The results showed that the weight of diabetic rats in each experimental group showed a decreasing trend, and the weight of rats in DMC group was significantly lower than that in DMC group and MTH group, and within 120 min after glucose administration, the weight of rats in LCM group was significantly lower than that in MTH group. The blood glucose levels in the LCM and HCM groups were significantly lower than those in the DMC group at four time points, while the fasting blood glucose levels and fasting insulin fins levels in the diabetic model rats were decreased by camel milk. After treatment with camel milk, some cells of liver returned to normal structure, the degree of steatosis was obviously reduced, there was no vacuolation, and the basic morphological structure of kidney was complete and clear. The expression of Bax and NF-?B protein was inhibited and the expression of Bcl-2 protein was up-regulated by camel milk. Compared with MTH, camel milk inhibited the expression of Bax and NF-?B m RNA, up-regulated the expression of caspase-3 m RNA, down-regulated the expression of Bcl-2 m RNA, and decreased the expression of Bax and NF-?B m RNA in kidney. The upregulation of Bcl-2 m RNA expression and the high expression of caspase-3 m RNA in LCM and HCM groups need further study. In conclusion, camel milk can improve physiological and biochemical indexes and inhibit the expression of apoptotic proteins and genes in liver and kidney tissues of type 2 diabetic rats. The results suggest that bactrian camel milk can protect the liver and kidney of type 2 diabetic rats.
