饮食诱导脂肪沉积对线虫产卵量的影响

【摘要】:随着社会发展,饮食结构也在发生改变,伴随着高营养物质的过量摄入,肥胖的比例在不断上升。与此同时,不孕不育的比例也在逐渐上升。由于营养对生殖发育具有重要影响,我们推测饮食诱导的脂肪沉积与生育率有重要影响。本研究利用模式生物线虫研究了上述问题,通过饮食添加营养物质,然后观察线虫的脂肪沉积和产卵量(brood size)变化,并研究了相关机制。首先调查了葡萄糖、胆固醇、棕榈酸、油酸四种营养物质对线虫产卵总量及脂肪沉积的影响。葡萄糖和棕榈酸添加后线虫的产卵总量下降,但是脂肪沉积增加。通过改变线虫的肠道菌群,我们研究了饮食添加葡萄糖、棕榈酸诱导线虫产卵总量和脂肪沉积表型的影响。用不同种类的大肠杆菌喂食线虫,同时添加葡萄糖和棕榈酸,调查产卵量和脂肪沉积表型。结果表明改变线虫的肠道菌群,添加葡萄糖或棕榈酸后线虫依然会出现产卵总量下降,脂肪沉积增加的表型。接下来我们探讨了饮食诱导线虫产卵总量下降的过程中,脂肪沉积改变的必要性。我们发现,葡萄糖、棕榈酸直接引起的线虫产卵总量的下降,不依赖脂肪沉积的改变;而胆固醇则可能是通过脂肪沉积增加而引起产卵量的下降。最后,我们探讨了调控上述现象的分子机制。通过定量PCR相关基因及突变体的研究,我们发现营养物质葡萄糖、棕榈酸诱导线虫产卵总量下降与蛋白质糖基化转移酶ogt-1和溶酶体组织蛋白酶cpl-1有关。综上所述,通过本文研究我们得出营养物质的添加会影响线虫产卵总量的改变,其中葡萄糖和棕榈酸添加情况下,诱导线虫产卵总量下降不依赖脂肪沉积的增加。而是通过蛋白质糖基化及溶酶体组织蛋白酶的作用。有利于研究由于饮食诱导肥胖导致繁殖力改变的相关研究、预防和治疗。
[Abstract]:With the development of the society, the diet structure is also changing, with the excessive intake of high nutrients, the proportion of obesity is increasing. At the same time, the proportion of infertility is increasing. Because of the important effect of nutrition on reproductive development, we speculate that the diet induced fat deposition and fertility have an important impact. The above problems were studied with model biological nematodes, and nutrients were added to the diet. Then the fatty deposits and brood size changes of nematodes were observed and the related mechanisms were studied. First, the effects of four nutrients, glucose, cholesterol, palmitic acid and oleic acid, on the amount of egg production and fat deposition of line worms. After the addition of the nematode, the total amount of spawning decreased, but the fat deposition increased. By changing the intestinal flora of the nematode, we studied the effects of dietary glucose, palmitic acid induced oviposition and the fat deposition phenotype. Feeding nematodes with different kinds of Escherichia coli, adding glucose and palmitic acid to investigate the amount of eggs and fat. The results showed that the nematode colony of the nematode was changed. After the addition of glucose or palmitic acid, the total amount of oviposition decreased and the fat deposition was increased. Then we explored the necessity of the change in the fat deposition in the process of reducing the total amount of oviposition of the nematode. We found that glucose and palmitic acid are directly caused. The decrease of the total amount of egg laying in the nematode is not dependent on the change in the fat deposition, and the cholesterol may be caused by the increase of the fat deposition. Finally, we explored the molecular mechanism to regulate the above phenomenon. Through the study of the quantitative PCR related genes and mutants, we found the nutrient substance glucose and the palmitic acid induced nematodes. The decrease of total egg production is related to the protein glycosyltransferase ogt-1 and lysosome cathepsin Cpl-1. In this paper, we conclude that the addition of nutrients will affect the change of the total amount of egg laying in the nematode. In the case of glucose and palmitic acid, the decrease of the total amount of oviposition of nematodes is not dependent on the increase of fat deposition. It is through protein glycosylation and lysosomal cathepsin. It is beneficial to study the related research, prevention and treatment of fecundity caused by diet induced obesity.
