不同海拔高度大鼠血浆中超氧化歧化酶、丙二醛水平变化

【摘要】:目的:通过建立三个不同海拔的动物实验模型,研究对照大鼠血浆中超氧化物歧化酶、丙二醛水平的变化,旨在探讨不同海拔对大鼠血浆中超氧化歧化酶活性、丙二醛含量变化的影响,为高海拔低氧环境下急慢性机体损伤的防治提供重要的理论基础。方法:24只雄性大鼠随机分为对照组(兰州,海拔1500米)、中度海拔组(西宁,海拔2260米)和高海拔组(低压氧舱,模拟海拔5000米)。将中度海拔组和高海拔组大鼠分别由兰州快速运至西宁地区和青海大学低压氧舱室,30天后抽取大鼠下腔静脉血标本,分别用水溶性四氮唑法测定超氧化歧化酶活力,硫代巴比妥酸比色法测定丙二醛含量。结果 :对照组大鼠血浆中超氧化歧化酶、丙二醛水平分别为(19.98±1.57)、(2.28±0.68),中海拔组血浆中超氧化歧化酶、丙二醛水平分别为(19.06±1.24)、(2.76±0.34),高海拔组大鼠血浆中超氧化歧化酶、丙二醛水平分别为(10.34±1.19)、(6.35±0.58)。高海拔组大鼠血浆中超氧化歧化酶活力明显高于中海拔组及对照组,丙二醛含量明显高于中海拔组及对照组(均P0.001);中海拔组大鼠血浆中超氧化歧化酶活力略低于对照组,高于模拟特高海拔组,丙二醛含量略高于对照组,低于模拟特高海拔组(均P0.001)。结论:在高海拔低氧条件下,大鼠血浆中超氧化歧化酶活力逐渐下降、丙二醛含量逐渐增加,提示机体抗氧化损伤能力减弱、机体细胞氧化损伤进行性发展、组织中自由基的含量进行性升高、由其导致的脂质过氧化反应程度越来越严重、机体细胞损伤的程度越来越严重。
[Abstract]:Objective: to study the changes of superoxide dismutase (SOD) and malondialdehyde (MDA) levels in plasma of control rats by establishing three animal models at different elevations in order to study the activity of superoxide dismutase (SOD) in plasma of rats at different elevations. The effect of malondialdehyde content provides an important theoretical basis for the prevention and treatment of acute and chronic organism injury in high altitude and low oxygen environment. Methods Twenty four male rats were randomly divided into control group (Lanzhou, 1500 meters above sea level), moderate altitude group (Xining, 2260 meters above sea level) and high altitude group (low oxygen chamber, simulated altitude 5000 meters). The blood samples of inferior vena cava of rats in moderate altitude group and high altitude group were collected after 30 days of rapid transport from Lanzhou to Xining area and low pressure oxygen chamber of Qinghai University. The activity of superoxide dismutase (SOD) was determined by water-soluble tetrazolium method. The content of malondialdehyde was determined by thiobarbituric acid colorimetry. Results: the levels of superoxide dismutase (SOD) and malondialdehyde (MDA) were (19.98 卤1.57), (2.28 卤0.68) in the control group, (19.06 卤1.24), (卤0.34) in the middle altitude group, and (10.34 卤1.19), (/ 6.35 卤0.58) in the high altitude group. The superoxide dismutase activity in plasma of rats in high altitude group was significantly higher than that in middle altitude group and control group, and the content of malondialdehyde in plasma was significantly higher than that in middle altitude group and control group (P0.001), and the activity of superoxide dismutase in plasma of middle altitude group was slightly lower than that of control group. The content of malondialdehyde was slightly higher than that of the control group and lower than that of the simulated super high altitude group (all P0.001). Conclusion: under the condition of high altitude hypoxia, the activity of superoxide dismutase (SOD) in plasma and the content of malondialdehyde (MDA) in plasma of rats were gradually decreased, which indicated that the ability of antioxidant damage was weakened and the oxidative damage of cells was gradually developed. The content of free radicals in tissues is increasing, the degree of lipid peroxidation is becoming more and more serious, and the degree of cell damage is becoming more and more serious.
