新疆汉族2型糖尿病患者miR-375基因启动子区甲基化的初步研究

【摘要】:目的探讨新疆汉族T2DM及糖耐量正常(NGT)人群miR-375启动子区部分CpG位点的甲基化状态。方法采用基质辅助激光解析电离时间飞行质谱仪检测100例T2DM患者(T2DM组)和100名NGT者(NGT组)外周血白细胞中miR-375基因启动子区5个CpG位点的甲基化状态,并对结果进行分析。结果 (1)T2DM组平均甲基化水平高于NGT组(10.27%vs 7.24%,P0.01);(2)T2DM组miR-375基因5个CpG位点的甲基化率均高于NGT组,如CpG_22、CpG_24.25、CpG_26.27,仅CpG_26.27在两组中比较,差异有统计学意义(P0.01)。结论汉族T2DM患者外周血白细胞中miR-375基因部分启动子区异常甲基化水平可能与T2DM的发生有一定的关系。
[Abstract]:Objective to investigate the methylation status of partial CpG loci in miR-375 promoter region of T2DM and normal glucose tolerance (NGT) population in Xinjiang Han nationality. Methods the methylation status of 5 CpG sites in peripheral blood leukocytes of 100 T2DM patients (T2DM group) and 100 NGT subjects (NGT group) was detected by matrix assisted laser desorption time flight mass spectrometer. The results were analyzed. Results (1) the average methylation level of miR-375 gene in T2DM group was higher than that in NGT group (10.27%vs 7.24g / P0.01); (2). The methylation rate of miR-375 gene in T2DM group was higher than that in NGT group. Conclusion the abnormal methylation level of partial promoter region of miR-375 gene in peripheral blood leukocytes of Han T2DM patients may be related to the occurrence of T2DM.
