硫辛酸对骨质疏松大鼠骨代谢的影响及机制研究
[Abstract]:Objective to investigate the role and possible mechanism of oxidative stress and antioxidant lipoic acid in the pathogenesis of osteoporosis in rats. Methods Twenty-four 8-week-old female Wistar rats were randomly divided into control group, osteoporosis group and lipoic acid intervention group. The model of osteoporosis was established by bilateral ovariectomy, and the rats in the lipoic acid group were intraperitoneally injected with lipoic acid for 60 mg/kg. Bone mineral density (BMD), whole blood alkaline phosphatase (ALP),) osteocalcin, calcium and phosphorus levels were measured for 8 weeks. The content of MDA-SOD and GSH-Px in femur tissue was determined by Western blotting. The expression of OPG and RANKL m RNA in femur tissue was determined by real-time PCR and (OPG) of osteoprotegerin and nuclear factor 魏 B receptor activating factor ligand (RANKL) protein were determined by real-time PCR. Results the bone mineral density (BMD), the expression of SODH-PxPxOPG m RNA and protein in femur in osteoporosis group were significantly lower than those in control group (P0.05), and the levels of serum osteocalcin (BGP) and MDA-RANKL m RNA and protein in femur were significantly higher than those in control group (P0.05). The bone mineral density (BMD) and the expression of SODH-PxPxOPG m RNA and protein in femur tissue of rats in lipoic acid group were significantly higher than those in osteoporosis group (P0.05), while the levels of serum osteocalcin and RNA and protein in femur tissue were significantly lower than those in osteoporosis group (P0.05). Conclusion oxidative stress may promote the development of osteoporosis in rats through OPG/RANKL pathway, and the antioxidant lipoic acid can alleviate the progression of osteoporosis.
