膜联蛋白A1对糖尿病大鼠心功能及炎症反应的影响

【摘要】:目的探讨膜联蛋白A1(Annexin A1)对糖尿病大鼠心功能及肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)的影响。方法将24只SD大鼠随机均分为正常对照组、糖尿病组;利用高糖高脂饮食饲养及较小剂量链脲佐菌素(30 mg/kg)诱导造成2型糖尿病模型,2组分别予以普通喂食、高糖高脂喂食;于第8周末用彩色多普勒超声检测左心室舒张末期容积(LVEDV)、左心室收缩末期容积(LVESV)、二尖瓣舒张早期峰值速度/二尖瓣舒张晚期峰值速度(e/a)比值、左心室射血分数(LVEF)、每搏射血量(SV);第0、4、8周采用实时PCR测定血中Annexin A1表达,采用酶联免疫分析法测定血中TNF-α、IL-1β。结果与正常对照组对比,糖尿病组LVEDV、e/a、SV明显下降,Annexin A1明显上升(P0.05),8周后糖尿病组Annexin A1显著下降(P0.01);与正常对照组对比,糖尿病组TNF-α、IL-1β上升明显(P0.05),8周后上升更明显(P0.01);糖尿病组Annexin A1与TNF-α、IL-1β负相关(r分别为-0.73、-0.71,P0.01)。结论 Annexin A1可改善糖尿病大鼠心功能,Annexin A1改善糖尿病大鼠心功能可能与炎症反应有关。
[Abstract]:Objective to investigate the effects of Annexin A1 on cardiac function and tumor necrosis factor 伪 (TNF- 伪) and interleukin 1 尾 (IL-1 尾) in diabetic rats. Methods Twenty-four Sprague-Dawley rats were randomly divided into normal control group and diabetic group, fed with high glucose and high fat diet and low dose streptozotocin (30 mg/kg). At the end of the 8th week, left ventricular end-diastolic volume (LVEDV), left ventricular end-systolic volume (LVEDV), early diastolic peak velocity (LVESV),) / end-diastolic peak velocity (E / a) ratio of left ventricular end diastolic volume (LVEDV),) were measured by color Doppler ultrasound. Left ventricular ejection fraction (LVEF),) was used to detect the expression of Annexin A1 in blood by real-time PCR and TNF- 伪 and IL-1 尾 by enzyme linked immunosorbent assay (Elisa). Results compared with normal control group, Annexin A1 decreased significantly in diabetic group (P0.05), and decreased significantly in diabetic group (P0.01), and decreased significantly in diabetic group (P0.05), and decreased significantly in diabetic group (P0.01) after 8 weeks, compared with normal control group. In diabetic group, the level of TNF- 伪 and IL-1 尾 increased significantly (P0.05) after 8 weeks (P0.01), and the correlation between TNF- 伪 IL-1 尾 and Annexin A1 was negative (r = -0.73 卤-0.71 P0.01, respectively). Conclusion Annexin A1 can improve cardiac function in diabetic rats.
