芦丁对肥胖诱发的雄性小鼠生殖损伤的保护作用

【摘要】:目的探讨芦丁在肥胖雄性小鼠体质量抑制以及对肥胖诱发的生殖损伤的保护效果及其机制。方法将24只雄性ICR小鼠随机分成3组,每组8只:正常对照组(ND组,正常饮食),高脂饮食组(HFD组,高脂饮食),芦丁保护组(HRU组,高脂饮食+芦丁)。28 d后,按后续实验需要进行取材处理。试剂盒检测生化指标;睾丸与附睾进行石蜡切片,HE染色进行形态学观察;统计精子数、活动度和畸形率。荧光定量PCR检测相关基因的表达水平;Western blot检测Ucp1蛋白表达水平。结果 HRU组比HFD组的平均体质量轻。HFD组睾丸和附睾组织中的甘油三酯的含量以及附睾组织中的甘油三酯的含量,明显高于ND组和HRU组。相较于HFD组的睾丸和附睾均出现结构松散,细胞结构异常,管腔内成熟的精子数量较少的现象,精子数量下降,活动度差等问题,HRU组均有明显改善。解偶联蛋白Ucp1基因和蛋白的表达量在芦丁处理后显著上升(P0.05),Mcp1和Tnfα的表达则显著下降(P0.05)。结论芦丁可以有效抑制高脂诱导的肥胖小鼠体质量的快速增长,对肥胖诱发的生殖损伤有一定的保护效果。
[Abstract]:Objective to investigate the protective effect and mechanism of rutin on body mass inhibition and reproductive injury induced by obesity in obese male mice. Methods Twenty-four male ICR mice were randomly divided into 3 groups: normal control group (ND group, normal diet), high-fat diet group (HFD group, high-fat diet) and rutin protection group (HRU group, high-fat diet rutin group). According to the need of subsequent experiments, material selection is carried out. Biochemical indexes were detected by kit, paraffin sections of testis and epididymis were detected, morphological observation was performed by HE staining, sperm count, mobility and deformity rate were counted. The expression level of related genes was detected by fluorescence quantitative PCR.; Western blot was used to detect the expression of Ucp1 protein. Results the average body weight of HRU group was lighter than that of HFD group. The contents of triglyceride in testis and epididymis in HFD group and in epididymal tissue were significantly higher than those in ND group and HRU group. Compared with the HFD group, the testis and epididymis showed loose structure, abnormal cell structure, small number of mature sperm in the lumen, decreased sperm number and poor mobility. The expression of uncoupling protein Ucp1 gene and protein increased significantly after rutin treatment (P0.05), while the expression of Mcp1 and Tnf 伪 decreased significantly (P0.05). Conclusion rutin can effectively inhibit the rapid growth of body mass in obese mice induced by high fat, and has a protective effect on the reproductive injury induced by obesity.
