吸烟对甲状腺相关眼病患者眶脂肪分化的影响

【摘要】:目的确定吸烟对甲状腺相关眼病(TAO)患者眶脂肪分化的影响。设计回顾性病例系列。研究对象2012年6-12月在上海长征医院眼科行眶减压术的TAO患者40例,男性27例,女性13例。方法 40例TAO患者中,21例吸烟、8例曾吸烟但已戒烟,5例被动吸烟,6例非吸烟。运用RT-PCR检测所有患者眶脂肪CCAAT/增强结合蛋白(C/EBP)和过氧化物酶体增生物激活受体(PPARγ)mRNA表达量。用△Ct法进行其各基因表达的相对定量。多组间的总体比较采用单因素方差分析,随后用SNK-q检验进行两两比较。主要指标C/EBP和PPARγmRNA表达量。结果吸烟组患者的C/EBP(12.04±4.26)和PPARγmRNA表达量(4.82±1.41)明显高于非吸烟组(C/EBP:2.32±0.92;PPARγ:1.37±0.25)(P0.001)。同时,曾吸烟组和被动吸烟组的C/EBP(4.29±1.26,5.76±1.03)和PPARγmRNA表达量(2.91±1.23,3.22±0.50)对比非吸烟组的差异也具有统计学意义(曾吸烟组C/EBP P=0.039,PPARγP=0.047;被动吸烟组C/EBP P=0.002,PPARγP=0.002)。吸烟指数≥100年支的TAO患者眶脂肪C/EBP(14.28±3.61)和PPARγmRNA(5.65±1.02)高于吸烟指数100年支的患者(C/EBP:8.42±2.30;PPARγ:3.48±0.74)(P均0.01)。结论眶脂肪C/EBP和PPARγmRNA的表达量与吸烟史有关。
[Abstract]:Objective to determine the effect of smoking on orbital adipose differentiation in patients with thyroid associated ophthalmopathy (TAO). Design retrospective case series. Participants 40 TAO patients, 27 males and 13 females, underwent orbital decompression in Shanghai long March Hospital from June to December 2012. Methods among 40 patients with TAO, 21 were smokers, 8 had smoked but had given up smoking, 5 were passive smokers and 6 were non-smokers. The expressions of CCAAT/ enhanced binding protein (C/EBP) and peroxisome proliferator-activated receptor (PPAR 纬) mRNA in orbital fat were detected by RT-PCR. The relative quantitative analysis of the gene expression was carried out by Ct method. Single factor analysis of variance (ANOVA) was used to compare the population of different groups, and then pairwise comparison was performed with SNK-q test. Main outcome measures: expression of C/EBP and PPAR 纬 mRNA. Results the expression of C/EBP and PPAR 纬 mRNA in smoking group (12.04 卤4.26) was significantly higher than that in non-smoking group (C/EBP:2.32 卤0.92) (1.37 卤0.25) (P0.001). At the same time, the expression of C/EBP and PPAR 纬 mRNA in the former smoking group and the passive smoking group were (4.29 卤1.26) 5.76 卤1.03 and (2.91 卤1.23) 3.22 卤0.50, respectively, which were significantly higher than those in the non-smoking group (C/EBP P0. 039 PPAR- 纬 P0. 047).
