肿瘤坏死因子-α诱导蛋白-8样分子-2在自身免疫疾病中的作用及意义

【摘要】：自身免疫疾病是机体对自身组织细胞产生异常免疫应答反应所导致的一类疾病,其确切发病机制仍未阐明。肿瘤坏死因子-α诱导蛋白-8样分子-2(TIPE2)是一种新近发现的免疫负性调控因子,属于肿瘤坏死因子-α诱导蛋白-8(TNFAIP8)家族成员。研究发现,TIPE2在多种自身免疫疾病中发生异常改变,并与疾病严重程度、病情进展及预后转归等密切相关,包括自身免疫性肝炎、原发性胆汁性肝硬化、重症肌无力、系统性红斑狼疮等,提示TIPE2在自身免疫疾病的发病中可能具有重要作用。TIPE2表达的缺陷可通过引起自身反应性T淋巴细胞或巨噬细胞的高反应性或促炎细胞因子大量释放入血,加重炎症反应并诱导自身免疫疾病的发生;或通过打破免疫稳态间接诱发自身免疫疾病。本文拟对TIPE2在自身免疫疾病中的作用与机制研究进展作一综述。
[Abstract]:Autoimmune diseases are a kind of diseases caused by abnormal immune response to their own tissues and cells. The exact pathogenesis of autoimmune diseases has not been clarified. Tumor necrosis factor-伪 inducible protein-8-like molecule-2 (TIPE2) is a newly discovered immuno-negative regulatory factor, which belongs to the tumor necrosis factor-伪 inducible protein-8 (TNFAIP8) family. It has been found that TIPE2 has abnormal changes in a variety of autoimmune diseases and is closely related to the severity, progression and prognosis of the disease, including autoimmune hepatitis, primary biliary cirrhosis, myasthenia gravis, and so on, including autoimmune hepatitis, primary biliary cirrhosis, myasthenia gravis, and so on. Systemic lupus erythematosus, et al., suggest that TIPE2 may play an important role in the pathogenesis of autoimmune diseases. The deficiency of TIPE2 expression may be caused by the high reactivity of autoreactive T lymphocytes or macrophages or the release of pro-inflammatory cytokines into the blood. Aggravate the inflammatory reaction and induce the occurrence of autoimmune diseases; Or indirectly induce autoimmune disease by breaking immune homeostasis. This paper reviews the role and mechanism of TIPE2 in autoimmune diseases.
