胰岛素对1型糖尿病大鼠睾丸脂联素及其受体、AMPK、AKT和eNOS表达的影响

【摘要】:目的:观察胰岛素干预1型糖尿病(T1DM)大鼠前后,睾丸组织中脂联素、脂联素受体及其介导的信号因子、炎性细胞因子的表达情况,探讨胰岛素对T1DM大鼠睾丸组织脂联素、脂联素受体及其介导的信号转导通路表达的影响。方法:30只清洁级SD雄性大鼠随机分成正常对照组(A组,n=8)和糖尿病造模组(n=22),造模组大鼠采用一次性腹腔注射65mg/Kg STZ以制备T1DM模型。将成模的16只雄鼠随机分为糖尿病对照组(B组,n=8)和糖尿病胰岛素治疗组(C组,n=8)。C组大鼠每日皮下注射精蛋白锌胰岛素3~5U/只,血糖控制于10.0mmol/L左右,A、B组大鼠予等量生理盐水。于实验8周末留取血标本,用于血生化指标、胰岛素(Ins.)及性激素(T、LH、FSH)水平检测,然后处死动物,迅速取出双侧睾丸及附睾,计数睾丸重量、精子数量及活动率,测定睾丸脂联素(Adiponectin)、脂联素受体(Adipo R)、AMP依赖的蛋白激酶(AMPK)、蛋白激酶B(AKT)、内皮型一氧化氮合酶(e NOS)、一氧化氮(NO)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的表达水平。统计采用LSD-t检验和单因素方差分析。结果:1.与A组比较,B组T1DM大鼠睾丸组织出现显著病理改变。2.与A组比较,B组T1DM大鼠的血糖、血脂、睾丸组织IL-6、TNF-α、P-AKT/AKT、e NOS和NO表达水平较A组显著升高(P0.05)。3.与A组比较,B组T1DM大鼠的血清Ins.和Adiponectin、血清和睾丸组织T、睾丸重量、精子数量及活动率、组织Adiponectin及Adipo R1、P-AMPK/AMPK水平较A组显著降低(P0.05)。4.胰岛素治疗能显著逆转上述各项指标的变化。结论:1.T1DM可导致大鼠睾丸组织发生严重病理改变。2.糖尿病大鼠病变睾丸组织中Adiponectin、Adipo R、信号转导因子AMPK、AKT、e NOS、NO及炎性细胞因子IL-6、TNF-α均发生相应改变,可能在T1DM大鼠睾丸组织病损中发挥了重要作用。3.胰岛素逆转了上述指标变化。胰岛素可能通过调节Adiponectin、Adipo R介导的信号转导通路减轻糖尿病大鼠睾丸组织损伤程度,发挥保护作用。
[Abstract]:Objective: to observe the expression of adiponectin, adiponectin receptor, signal factor and inflammatory cytokines in testis of type 1 diabetes mellitus (T1DM) rats before and after insulin intervention, and to explore the effect of insulin on adiponectin in testis of T1DM rats. Effects of adiponectin receptor and its signal transduction pathway on the expression of adiponectin receptor. Methods: thirty clean grade SD male rats were randomly divided into two groups: normal control group (group A, n = 8) and diabetic model group (n = 22). T1DM model was made by intraperitoneal injection of 65mg/Kg STZ at one time in the model group. Sixteen male rats were randomly divided into two groups: diabetic control group (group B, n = 8) and diabetic insulin treatment group (group C, n = 8). C). The rats were subcutaneously injected with protamine zinc insulin (3~5U/) every day, and the blood glucose was controlled around 10.0mmol/L, A. Group B rats were given the same amount of saline. Blood samples were collected at the end of experiment 8 to be used as biochemical index of blood, insulin (Ins.) The levels of sex hormones (T, LH, FSH) were measured, and then the animals were killed. Bilateral testes and epididymis were taken out quickly. The weight of testis, sperm count and motility were counted. The adiponectin (Adiponectin), adiponectin receptor (Adipo R), in testis was determined. Expression of AMP-dependent protein kinase (AMPK), protein kinase B (AKT), endothelial nitric oxide synthase (e NOS), nitric oxide (NO), interleukin-6 (IL-6) and tumor necrosis factor-伪 (TNF- 伪). LSD-t test and one-way ANOVA were used. Results: 1. Compared with group A, there were significant pathological changes in testicular tissue of T1DM rats in group B. Compared with group A, the levels of blood glucose, blood lipid, IL-6,TNF- 伪, pAKT / Akt, e-NOS and NO in T1DM rats in group B were significantly higher than those in group A (P0.05). Serum Ins. of T1DM rats in group B compared with group A And Adiponectin, serum and testicular tissue T, testicular weight, sperm quantity and motility, tissue Adiponectin, Adipo R1, PxAMPK and AMPK levels were significantly lower than those in group A (P0.05). Insulin therapy can significantly reverse the changes of the above indexes. Conclusion: 1.T1DM can cause severe pathological changes in testicular tissue of rats. The changes of Adiponectin,Adipo R, signal transduction factor AMPK,AKT,e NOS,NO and inflammatory cytokine IL-6,TNF- 伪 in testicular tissue of diabetic rats may play an important role in the pathological changes of testicular tissue in T1DM rats. 3. Insulin reversed these changes. Insulin may play a protective role by modulating the signal transduction pathway mediated by Adiponectin,Adipo R to reduce the degree of testicular injury in diabetic rats.
