肝细胞生长因子调节的酪氨酸激酶底物对胰岛素表达和分泌的影响

【摘要】:目的探讨肝细胞生长因子调节的酪氨酸激酶底物(HGS)对胰岛素表达和分泌的影响。方法检测HGS,胰岛素和胰升血糖素在小鼠胰岛中的分布。检测不同浓度葡萄糖(5.5、11.1及16.7mmol/L)培养24h后NIT-1细胞HGS表达水平和胰岛素的分泌。观察siRNA沉默HGS表达后48h NIT-1细胞胰岛素的表达和分泌的变化。结果 HGS免疫阳性细胞特异性分布于小鼠胰腺的胰岛,并与胰岛素共定位于胰岛β细胞中,未见HGS和胰高血糖素阳性荧光双标细胞。不同浓度葡萄糖培养NIT-1细胞24h,5.5mmol/L组为(25.07±0.93)μIU/mg,11.1mmol/L组为(32.28±0.93)μIU/mg,16.7mmol/L组为(41.77±1.39)μIU/mg,NIT-1细胞分泌胰岛素水平逐渐增加(P0.01)。各组HGS蛋白水平也随葡萄糖浓度增加而逐渐增加(P0.05)。siRNA沉默HGS表达后NIT-1细胞内胰岛素的表达水平无明显变化,但胰岛素分泌水平减少(P0.05)。结论 HGS选择性表达于胰岛β细胞中,并调节胰岛素的分泌。
[Abstract]:Objective to investigate the effect of hepatocyte growth factor regulated tyrosine kinase substrate (HGS) on insulin expression and secretion. Methods the distribution of HGS, insulin and glucagon in mouse islets was detected. The expression of HGS and insulin secretion in NIT-1 cells cultured with different concentrations of glucose (5.5, 11.11 and 16.7mmol/L) for 24 h were detected. To observe the changes of insulin expression and secretion in NIT-1 cells 48 hours after siRNA silencing HGS expression. Results HGS immunopositive cells were specifically distributed in the islets of mouse pancreas and co-located in islet 尾 cells with insulin. No HGS and glucagon positive fluorescent double labeled cells were found. NIT-1 cells cultured at different concentrations of glucose for 24 hours were (25.07 卤0.93) 渭 IU/mg,11.1mmol/L and (31.77 卤1.39) 渭 IU/mg, in 5.5 mmol / L group, (32. 28 卤0. 93) 渭 IU/mg,16.7mmol/L group and (41.77 卤1.39) 渭 IU/mg,. The level of insulin secreted by NIT-1 cells increased gradually (P 0.01). The level of HGS protein in each group also increased gradually with the increase of glucose concentration (P 0.05). After silencing HGS expression, the expression level of insulin in NIT-1 cells did not change significantly, but the level of insulin secretion decreased (P 0.05). Conclusion HGS is selectively expressed in islet 尾 cells and regulates insulin secretion.
