茶多酚对果糖诱导的高尿酸血症大鼠血尿酸水平的影响及机制

【摘要】:目的:观察茶多酚(green tea polyphenols,GTP)对果糖诱导的高尿酸血症(hyperuricemia,HUA)大鼠血尿酸水平的影响,并从调节尿酸产生和排泄途径探讨其药理机制。方法:连续给予大鼠10 g/100 mL果糖溶液8周,其中5~8周分别连续灌胃别嘌呤醇(4 mg/(kg·d))或GTP(75、150、300 mg/(kg·d)),每天1次。磷钨酸法检测血尿酸水平,比色法和Western blot法分别检测肝脏黄嘌呤氧化酶(xanthine oxidase,XOD)活力及其表达水平,免疫组织化学染色法检测肾脏中尿酸盐转运子(urate transporter,UAT)、尿酸盐阴离子转运子(urate-anion transporter,URAT)1和葡萄糖转运子(glucose transporter,GLUT)9的表达水平。结果:150、300 mg/(kg·d)GTP显著降低了果糖诱导的HUA大鼠血尿酸水平(P0.05,P0.01)。同时,150、300 mg/(kg·d)GTP显著降低了HUA大鼠肝脏XOD活力和表达水平(P0.05,P0.01),显著增强了肾脏UAT表达水平和降低了肾脏URAT1和GLUT9的表达水平(P0.05,P0.01)。结论:150、300 mg/(kg·d)GTP可有效降低果糖诱导的HUA大鼠血尿酸水平,其机制与减少尿酸产生和促进尿酸排泄密切相关。
