补体C3f去精氨酸片段对系统性硬化症小鼠皮肤纤维化模型构建及转化生长因子β1表达的影响

【摘要】:目的探讨补体C3f去精氨酸片段(DRC3f)对于系统性硬化症(SSc)小鼠皮肤纤维化模型构建和转化生长因子(TGF)-β1表达的影响。方法采用DRC3f联合博来霉素皮下注射的方法构建SSc小鼠模型,并用纤维染色和免疫组化方法比较联合与不联合DRC3f对SSc小鼠皮肤纤维化、皮肤增厚程度和TGF-β1表达的影响。结果联合DRC3f和博来霉素注射组与单用博来霉素和博来霉素联合无关多肽组比较,皮肤纤维化程度更加明显,TGF-β1的表达也明显增多(P0.05)。结论 DRC3f能够促进SSc模型小鼠皮肤纤维化和炎症因子TGF-β1的表达。
[Abstract]:Objective to investigate the effect of complement C3f dearginine fragment (DRC3f) on the construction of skin fibrosis model and the expression of transforming growth factor (TGF)-尾 1 in (SSc) mice with systemic sclerosis. Methods SSc mouse model was established by DRC3f combined with bleomycin subcutaneous injection. The effects of DRC3f on skin fibrosis, skin thickening and TGF- 尾 1 expression in SSc mice were compared by fiber staining and immunohistochemistry. Results compared with the control group, the degree of skin fibrosis was more obvious and the expression of TGF- 尾 1 was significantly increased in the combined DRC3f and bleomycin injection group (P 0.05). Conclusion DRC3f can promote skin fibrosis and the expression of inflammatory factor TGF- 尾 1 in SSc model mice.
