IGFBP2通过ERK1/2信号通路调控RA-FLS细胞生物学行为机制研究

  目的:探讨胰岛素样生长因子结合蛋白2(Insulinlike growth factor binding protein2,IGFBP2)对类风湿关节炎成纤维样滑膜细胞(RA-FLS)生物学行为的作用,以及IGFBP2对细胞外调节蛋白激酶(Extracellular regulated protein kinases1/2,ERK1/2)的作用。方法:首先,我们原代培养类风湿关节炎成纤维样滑膜细胞(RA-FLS)和正常成纤维样滑膜细胞(FLS),并采用q RT-PCR、Western blot及免疫荧光技术检测IGFBP2 m RNA和蛋白在FLS和RA-FLS中的表达水平。其次,RA-FLS细胞被转染si RNA-IGFBP2和si RNA-NC,q RT-PCR检测IGFBP2在RA-FLS细胞中的干扰效果;CCK-8实验检测IGFBP2敲低后对RA-FLS细胞增殖能力的影响;流式细胞仪检测IGFBP2敲低后对RA-FLS细胞凋亡的影响;Transwell检测IGFBP2敲低对RA-FLS细胞侵袭能力的影响;划痕实验检测IGFBP2敲低对RA-FLS细胞迁移能力的影响。此外,RA-...
