利拉鲁肽对高糖诱导人髓核细胞调亡的作用和对2型糖尿病患者血糖变异性影响

胰高血糖素样肽-1(Glucagon-like peptide-1,GLP-1)是一种重要的肠促胰素,主要由肠道L细胞合成并分泌,葡萄糖依赖性促进胰岛β细胞分泌胰岛素,抑制胰岛α细胞分泌胰高血糖素。除降糖作用外,GLP-1通过影响迷走神经传入纤维和下丘脑,抑制食欲并延缓胃排空,从而减轻体重。GLP-1通过与GLP-1受体(GLP-1 receptor,GLP-1R)结合发挥作用。在生理状态下,循环中的GLP-1被二肽基肽酶IV(dipeptidyl peptidase IV,DPP-IV)迅速降解。利拉鲁肽是GLP-1受体激动剂(GLP-1 receptor agonists,GLP-1RA)的代表药物之一,与内源性GLP-1结构具有97%同源性,能有效抵抗DPP-IV的降解,且具有更长的半衰期,目前被认为是治疗2型糖尿病强有力的药物选择。糖尿病是椎间盘退变的潜在病因之一。糖尿病患者椎间盘退变的发病率高于非糖尿病患者。高糖对椎间盘细胞的生物学产生负面影响,诱导椎间盘细胞衰老和凋亡。由于两者之间的相关性,降糖药物对椎间盘退变的影响受到越来越多的关注。除了对血糖控制有益之外,利拉鲁肽在多种...
