血管内皮生长因子及其受体1和受体2在皮肤鳞状细胞癌及基底细胞癌中的表达

【摘要】:目的 皮肤鳞状细胞癌(squamous cell carcinoma, SCC)以及基底细胞癌(basal cell carcinoma, BCC)同源于表皮,共属于非黑色素性肿瘤。临床研究证实,SCC转移率较高,预后差,而BCC主要表现为局部破坏性浸润,极少发生转移,这两种肿瘤生物学行为有较大差异。 目前很多研究都证明了VEGF及其受体在多数实体肿瘤组织中表达明显增高,但是目前关于VEGFR-2在SCC中的表达,及VEGFR-1与VEGFR-2在SCC与BCC组织中表达的差异和相关性研究目前鲜有文献报道。本研究利用免疫组化SP法,定量检测SCC, BCC及正常皮肤组织中VEGF, VEGFR-1, VEGFR-2三种蛋白的表达,并比较其之间的差异性以及SCC组织中三种蛋白表达的相关性,来探讨三者在非黑色素性皮肤癌的发生中所起的作用,从而为皮肤肿瘤的防治提供新的思路。 方法 收集手术切除的皮肤鳞状细胞癌石蜡包埋标本26例,其中高分化16例,中低分化10例;皮肤基底细胞癌标本18例;取10例正常皮肤为对照组。采用免疫组化SP法检测其中VEGF, VEGFR-1, VEGFR-2的表达,三种蛋白的表达采用积分光密度值进行分析,用已知的VEGF, VEGFR-1, VEGFR-2阳性的皮肽鳞状细胞癌及堆底细胞癌标本做为阳性刈‘j!{{,以PBS代替‘抗孵育组织切片作为阴性对照;结合临床病理分级,应用SPSS17.0统计软件进行统计学分析。 结果 1. VEGF, VEGFR-1, VEGFR-2主要表达J:JJIll瘤实质细J胞胞浆,农皮』-I底层以上除角质层外的细胞,部分血管内J皮细胞可见5j5j表达,为胞质Il]棕黄色’染色颗粒。 2. VEGF在鳞状细胞癌,基底细胞癌以及难常J皮肤·l]的秋分光街度值分别为(129.052±9.933),(116.067±3.864)以及(85.254±12.982),VEGFR-1在其三者中的表达水平为(130.481±12.020),(117.453±8.157)以及(82.193±9.150),VEGFR-2分别为(136.604±6.672),(121.535±7.499)以及(85.430±9.334)。VEGF, VEGFR-1, VEGFR-2在SCC组织[lll的阳性表达与BCC及IIi常皮肤组的表达比较,均有显著性差异(F=83.418,78.137以及169.125,P0.05),且征SCC组织中表达显著i盏于BCC(P0.05);BCC LflVEGF, VEGFR-1, VEGFR-2的表达与正常皮肽相比,差异有统计学意义(P0.05)。 3. VEGF在i高分化及中低分化鳞状细胞癌中的积分光密度值分别为(124.492±8.969),(136.346±6.630):VEGFR-1征高分化及lll低分化鳞状细胞癌中的积分光密度值分别为(127.076±13.616),(135.930±6.221);VEGFR-2在高分化及中低分化鳞状细胞癌中的积分光密度值分别为(133.573+_.6.563),(141.453±3.065)。VEGF及VEGFR-2在tIIf氐分化SCC中的表达显著高于高分化SCC(P0.05),即分化越差,表达越高;高分化与中低分化SCC中的VEGFR-1表达比较,差异不具统计学意义(P0.05)。 4.在皮肤鳞状细胞癌11],VEGF的表达程度与VEGFR-1成正相关1(r=0.773,P0.05),IjVEGFR-2的表达成II渊I关(r=0.676,P0.05),.IJVEGFR-1的表达水j川jVEGFR-2成_II渊j关(r=0.718,P0.05)。 结论 1.皮肤鳞状细胞癌和基底细胞煽中,VEGF, VEGFR-1, VEGFR-2的蛋白表达水平可能。¨C生物’学特征相关, i符-f门异常斯表达IIJ‘能为非黑色索性J皮肤癌过增殖的原因之。 2. VEGF, VEGFR-2的阳性表达与SCC病理学分级呈负相关,而VEGFR.1的阳性表达与SCC组织学分级无明显关系。 3. VEGF, VEGFR-1, VEGFR-2蛋白在SCC中的表达呈正相关,三者可能共同促进肿瘤的血管生成。 4. VEGF, VEGFR-1, VEGFR-2在皮肤鳞状细胞癌和基底细胞癌的发生发展过程中可能起着重要作用,联合检测三者在非黑色素性皮肤癌中的表达可能作为肿瘤的诊断,预后判断标准之一。
[Abstract]:objective
Cutaneous squamous cell carcinoma (squamous cell, carcinoma, SCC) and basal cell carcinoma (basal cell, carcinoma, BCC) homologous to the epidermis, belong to non melanoma tumors. Clinical studies have confirmed that SCC transfer rate is high, the prognosis is poor, while BCC is mainly manifested as local destructive infiltration, fewer metastases, two the biological behavior of the tumor are quite different.
At present, many studies have demonstrated that the expression of VEGF and its receptor in most solid tumor tissues were significantly increased, but the expression of VEGFR-2 in SCC in the study of differences and correlation between the expression of VEGFR-1 and VEGFR-2 in SCC and BCC tissues and there are few reports. This study used immunohistochemical SP method for quantitative detection of SCC. VEGF, BCC and VEGFR-1 in normal skin tissues, the expression of VEGFR-2 three protein, and three protein expression differences between the correlation and the SCC organization, to explore the three plays in the event of non melanoma skin cancer in the role, to provide new ideas for prevention and treatment of skin cancer.
Method
Cutaneous squamous cell carcinoma paraffin embedded specimens were collected from surgical operation in 26 cases, including 16 cases of high differentiation, 10 cases of low differentiated specimens; basal cell carcinoma in 18 cases; 10 cases of normal skin as control group. Immunohistochemical SP method was used to detect the VEGF, VEGFR-1, VEGFR-2 expression, expression of the three proteins the integral optical density values were analyzed using known VEGF, VEGFR-1, VEGFR-2 positive skin squamous cell carcinoma and basal cell carcinoma specimens as a heap of positive mow "J! PBS" instead of turning, with anti incubated tissue slices as negative control; combined with the clinical pathological grade, were analyzed by statistics software SPSS17.0.
Result
1. VEGF, VEGFR-1, VEGFR-2 mainly expressed J:JJIll cell parenchyma, fine J cytoplasm, agricultural skin, -I above the bottom layer, except for stratum corneum, some J cells in blood vessels showed 5j5j expression, cytoplasm Il] brown yellow staining granules.
2. VEGF in squamous cell carcinoma, basal cell carcinoma and often difficult to J l] street light skin equinox degree = (129.052 + 9.933), (116.067 + 3.864) and (85.254 + 12.982), the expression level of VEGFR-1 in three of the (130.481 + 12.020), (117.453 + 8.157) to and (82.193 + 9.150) VEGFR-2, respectively (136.604 + 6.672), (121.535 + 7.499) and (85.430 + 9.334).VEGF, VEGFR-1, compare the expression of VEGFR-2 in the positive expression of BCC and IIi and SCC [lll normal skin group, there were significant differences (F=83.418,78.137, 169.125, P0.05, SCC) and the I lamp syndrome expression was significantly in BCC (P0.05); BCC LflVEGF, VEGFR-1, VEGFR-2 expression was compared with normal skin, the difference was statistically significant (P0.05).
The integral optical density of 3. VEGF I in high differentiation and low differentiation squamous cell carcinoma respectively (124.492 + 8.969), (136.346 + 6.630):VEGFR-1 syndrome integral optical density high differentiation and low differentiation of LLL squamous cell carcinoma respectively (127.076 + 13.616), (135.930 + 6.221); integral light the density of VEGFR-2 in high differentiation and low differentiation squamous cell carcinoma respectively (133.573+_.6.563), (141.453 + 3.065) the expression of.VEGF and VEGFR-2 in tIIf di SCC differentiation was significantly higher than that of high differentiated SCC (P0.05), the poorer the differentiation, the higher the expression; expression of high differentiation and low differentiation in SCC VEGFR-1, the difference was not statistically significant (P0.05).

conclusion
1. skin squamous cell carcinoma and basal cell fans, VEGF, VEGFR-1, the expression level of VEGFR-2 protein may be "C". The biological characteristics of I, -f, abnormal expression of IIJ with the door "for non black J simply because of the proliferation of skin cancer too.
2. VEGF, the positive expression of VEGFR-2 was negatively correlated with the pathological grade of SCC, but the positive expression of VEGFR.1 was not significantly related to the histological grade of SCC.
The expression of 3. VEGF, VEGFR-1, and VEGFR-2 protein in SCC is positively correlated, and the three may jointly promote the angiogenesis of the tumor.
4. VEGF, VEGFR-1 and VEGFR-2 may play an important role in the occurrence and development of cutaneous squamous cell carcinoma and basal cell carcinoma. Combined detection of three expression in non melanoma skin cancer is probably one of the criteria for the diagnosis and prognosis of tumors.
