P物质在慢性自发性荨麻疹嗜碱性粒细胞中表达上调及其对组胺释放和嗜碱性粒细胞募集的诱导作用

【摘要】:目的通过检测慢性自发性荨麻疹患者外周血中嗜碱性粒细胞SP和NK1R的表达,以及SP对嗜碱性粒细胞的生物学功能和细胞募集的影响,阐明SP和嗜碱性粒细胞在慢性自发性荨麻疹的发生发展中可能存在的作用,为理解慢性自发性荨麻疹的发病机制提供新思路,为治疗慢性自发性荨麻疹提供新靶点。方法在本研究中,我们使用流式细胞仪分析检测了慢性自发性荨麻疹患者外周血嗜碱性粒细胞的SP和NK1R的表达以及嗜碱性粒细胞的数目;通过嗜碱性粒细胞激发试验研究SP对嗜碱性粒细胞组胺释放的影响;通过小鼠致敏模型的实验方法分别检测了小鼠外周血中嗜碱性粒细胞SP和NK1R的表达以及SP对嗜碱性粒细胞的功能影响。结果在CSU患者血浆SP水平和嗜碱性粒细胞数分别比HC受试者??更高。与HC受试者的外周血比较,CSU患者中SP+和NK1R+嗜碱性粒细胞的比例均显著升高。加入SP后,可引发CSU患者嗜碱性粒细胞高达41.2%的净组胺释放率,其促进作用甚至可与抗Ig E和f MLP的相比较。腹腔注射以后,SP可引发小鼠外周血嗜碱性粒细胞数目的显著增加。OVA致敏小鼠比非致敏小鼠外周血具有更多SP+和NK1R+嗜碱性粒细胞。结论高水平的血浆SP浓度,高表达的嗜碱性粒细胞SP和NK1R,以及SP的诱导嗜碱性粒细胞脱颗粒和募集的能力,都强有力的表明,SP是一种潜在的促炎性因子。它通过作用于嗜碱性粒细胞在CSU的发病机制中发挥重要作用。SP的抑制剂和NK1R的阻断剂可能会成为治疗CSU的有效制剂。
[Abstract]:Objective to investigate the expression of basophil SP and NK1R in peripheral blood of patients with chronic spontaneous urticaria and the effects of SP on the biological function and cell recruitment of basophil. To elucidate the possible role of SP and basophil in the occurrence and development of chronic spontaneous urticaria and to provide new ideas for understanding the pathogenesis of chronic spontaneous urticaria. Methods in this study, we used flow cytometry to detect the expression of SP and NK1R and the number of basophil in peripheral blood of patients with chronic spontaneous urticaria. The effect of SP on histamine release from basophil was studied by basophil stimulation test. The expression of SP and NK1R in peripheral blood of mice and the effect of SP on the function of basophil were detected by mouse sensitization model. Results in patients with CSU, plasma SP level and basophil were detected. More than HC subjects? ? Compared with the peripheral blood of HC subjects, the percentage of SP and NK1R eosinophils in patients with CSU was significantly higher than that in patients with HC.After the addition of SP, the net histamine release rate of basophil was up to 41.2% in patients with CSU. After intraperitoneal injection, SP could induce a significant increase in the number of basophil in peripheral blood of mice. OVA-sensitized mice had more SP and NK1R eosinophils than non-sensitized mice. Conclusion the high level of plasma SP concentration, Overexpression of basophil SP and NK1R, and SP's ability to induce degranulation and recruitment of basophil, It is strongly suggested that SP is a potential proinflammatory factor, which may be an effective agent for the treatment of CSU by acting on basophil. The inhibitor of SP and the antagonist of NK1R may play an important role in the pathogenesis of CSU by acting on basophilic granulocytes.
