特应性皮炎患者microRNAs表达的初步研究

【摘要】：目的：检测特应性皮炎(Atopic dermatitis, AD)患者外周血单个核细胞（Peripheral Blood Mononuclear Cells, PBMCs）中miR-155和miR-142-5p的表达情况，，为进一步研究miR-155、miR-142-5p在特应性皮炎发生、发展中的作用提供理论依据。材料与方法：收集8例AD患者及6例正常人外周静脉血各5ml，人淋巴细胞分离液分离PBMCs，miRNA提取分离试剂盒提取分离PBMCs中的miRNA,采用实时荧光定量PCR法，检测AD患者和正常人PBMCs中miR-155、miR-142-5p的表达水平。以U6snRNA为内参，采用2-ΔΔCT法计算miR-155、miR-142-5p在AD患者PBMCs中的相对表达量，分析AD的严重程度（SCORAD）评分与miR-155、miR-142-5p的表达水平之间的关系。用SPSS17.0统计软件对实验数据进行统计学分析，以α=0.05作为检验标准。结果：1. miR-155、miR-142-5p在AD患者PBMCs中均有表达。2.与正常对照组相比，miR-155、miR-142-5p在AD患者PBMCs中的表达水平上调（p＜0.05）。3. miR-155的相对表达量与AD严重程度积分有相关倾向(r=0.695, p=0.056),而miR-142-p的相对表达量与其无相关性(r=0.283, p=0.497)。结论：1.特应性皮炎患者PBMCs中存在miR-155、miR-142-5P，并可以用实时荧光定量PCR进行检测。2. miRNA-155、miRNA-142-5p在AD患者PBMCs中表达上调，可能在AD的发生、发展过程中发挥一定作用。3. miR-155的相对表达量与AD严重程度积分有相关倾向,而miR-142-p的相对表达量与其无相关性。
[Abstract]:Objective: to detect the expression of miR-155 and miR-142-5p in peripheral Blood Mononuclear cells (PBMCs) in patients with atopic dermatitis (AD), in order to further study the occurrence of miR-155 miR-142-5p in atopic dermatitis. Materials and methods: 5 ml of peripheral venous blood were collected from 8 patients with AD and 6 cases of normal controls. The isolation kit was used to extract miRNAs from peripheral blood of 8 patients with AD and 6 normal controls. Real time fluorescence quantitative PCR method was used to extract miRNAs from PBMCs. The expression of miR-155 miR-142-5p was detected in AD patients and normal subjects, and the relative expression of miR-155 miR-142-5p in PBMCs of AD patients was calculated by 2- 螖 CT using U6snRNA as internal control. The relationship between the severity of AD and the expression level of miR-155 miR-142-5p was analyzed. The experimental data were statistically analyzed by SPSS17.0 software. Results: 1. MiR-155 miR-142-5p was expressed in PBMCs of AD patients. The expression level of miR-155 miR-142-5p was up-regulated in PBMCs of AD patients compared with normal controls. The relative expression of miR-155 was significantly correlated with the score of AD severity. Conclusion: 1. MiR-155 miR-142-5P exists in PBMCs of patients with atopic dermatitis, and can be detected by real-time fluorescence quantitative PCR to detect upregulation of .2miRNA-155 miRNA-142-5p in PBMCs of AD patients. It may play a certain role in the occurrence and development of AD. The relative expression of miR-155 has a tendency to correlate with the score of AD severity, but the relative expression of miR-142-p has no correlation with AD.
