祛斑方对紫外线照射后色素沉着动物模型病理形态学影响

【摘要】:目的:探讨祛斑方对紫外线照射后色素沉着豚鼠模型的病理形态学影响。方法:采用中波紫外线照射白色雄性豚鼠复制黄褐斑实验动物模型,用免疫组化法检测祛斑方对皮肤黑素细胞的影响,并通过透射电镜法观察黑素小体的表达。结果:1黑色素阳性细胞的表达:造模后,模型组与空白组比较黑色素阳性细胞的数量显著增多(P0.05),与模型组比较,3个给药组黑色素阳性细胞数量明显下降(P0.05);祛斑方高剂量组显著优于西药对照组(P0.05)。2免疫组化染色病理切片结果:空白组表皮可见到极少个黑素细胞,模型组表皮层增厚,表皮基底细胞和棘细胞层中可见多个黑素细胞,呈强阳性反应;祛斑方低、高剂量组表皮基底细胞和棘细胞层中可见一些散在分布的黑素细胞;西药对照组表皮基底细胞和棘细胞层中可见数个散在分布的黑素细胞。3电子显微镜观察结果:空白组黑色素细胞细胞浆丰富,含有极少量直径小的黑素小体,模型组视野中可见表皮细胞中存在较多Ⅲ、Ⅳ期黑素小体,祛斑方高、低剂量组视野内黑素小体、黑素生成均较少,西药对照组细胞核上方可见数个黑素小体,直径较小。结论:祛斑方对紫外线照射后色素沉着动物模型具有良好的治疗作用,其机制可能是通过抑制黑素细胞的增殖,降低黑素细胞的功能达到的。
[Abstract]:Objective: to study the pathomorphological effect of Qubanfang on pigmented guinea pig model after ultraviolet irradiation.Methods: the experimental animal model of chloasma was established by ultraviolet B irradiation in white male guinea pigs. The effect of Qubanfang on melanocytes was detected by immunohistochemical method, and the expression of melanosome was observed by transmission electron microscope (TEM).Results the expression of melanin positive cells: after modeling,Compared with the blank group, the number of melanin positive cells in the model group was significantly higher than that in the blank group. Compared with the model group, the number of melanin positive cells in the three administration groups was significantly lower than that in the control group, and that in the high dose group was significantly better than that in the western medicine control group.The results of staining pathological sections showed that very few melanocytes could be seen in the epidermis of blank group.In the model group, many melanocytes were found in epidermal basal cell and spinous cell layer, and some scattered melanocytes were found in epidermal basal cell and spinous cell layer in high dose group.In the epidermal basal cell and spinous cell layer of the control group, the results of electron microscope observation showed that the melanocytes in the blank group were rich in cytoplasm and contained a very small number of melanosomes with small diameters.In the model group, there were more 鈪,
