紫外线诱导裸鼠皮肤鳞癌动物模型的构建

【摘要】:[目的]通过紫外线投照诱导裸鼠皮肤鳞癌,建立与人体头面部皮肤鳞癌相似的动物模型,探索投照过程中所需紫外线的投照波长、剂量、时间。 [方法]取35只健康的Nu/Nu裸鼠、雌雄不限、3周龄、体重10g左右,随机将其分为7组,给予不同波长、剂量的紫外线投照,分别为空白对照组、UVA(3.6J/cm2/day)、UVA(7.2 J/cm2/day)、UVB(720 mJ/cm2/day)、UVB(1440mJ/cm2/day)、UVA+UVB(3.6 J/cm2/day+720 mJ/cm2/day)、UVA+UVB(7.2 J/cm2/day+1440 mJ/cm2/day)。定期肉眼观察裸鼠背部皮肤的变化,照射4周、8周、12周、16周后定时切取裸鼠背部局部病变皮肤组织,行常规病理检查和免疫组织化学染色观察裸鼠背部皮肤变化过程,分析皮肤组织中上皮角蛋白CK10、CK5/6表达强度,通过SPSS11.0软件包采用单因素方差分析方法对其在不同时间位点的表达强度进行统计学分析。 [结果]空白对照组、UVA(3.6 J/cm2/day)、UVA(7.2 J/cm2/day)组肉眼观察裸鼠皮肤无明显异常变化,HE染色皮肤上皮层无明显异常变化。UVB(720mJ/cm2/day)及UVA+UVB(3.6 J/cm2/day+720 mJ/cm2/day)组照射16周后裸鼠背部皮肤不断增生、破溃,HE染色未见明显癌变,上皮层表现为非典型增生、大量炎性细胞浸润或局部溃疡。UVB(1440mJ/cm2/day)及UVA+UVB(7.2 J/cm2/day+1440 mJ/cm2/day)照射16周后裸鼠背部皮肤明显增生、破溃,形成火山口状或乳头状增生HE染色表现为高分化鳞癌。CK5/6、CK10在UVA照射组中的表达强度和正常裸鼠皮肤基本一致,在UVB或UVA+UVB照射组的表达随着照射剂量的增强、照射时间的增加,其阳性表达率逐渐增加。 [结论]①紫外线中的UVB是引起皮肤鳞癌的主要原因。②裸鼠皮肤鳞癌的发生与紫外线的投照波长、剂量及时间有关。③CK5/6、CK10在紫外线致皮肤损伤癌变的不同时间段表达强度不同,随着照射剂量的增强、照射时间的增加,其阳性表达率逐渐增加。
[Abstract]:[objective] to establish an animal model similar to that of human head and face skin squamous cell carcinoma (SCC) induced by ultraviolet radiation in nude mice, and to explore the ultraviolet radiation wavelength, dose and time. [methods] Thirty-five healthy Nu/Nu nude mice were randomly divided into 7 groups. They were randomly divided into 7 groups. They were exposed to ultraviolet rays of different wavelengths and dosages, respectively, as the blank control group: UVA3.6J / cm ~ (-2) / cm ~ (2) J / cm ~ (2) / day UVB 720mJ / cm ~ (-1) 路cm ~ (-2) / day UVA UVB(3.6 J/cm2/day 720mJ / cm ~ (2) / day UVA UVB(7.2 J/cm2/day 1440 mJ / cm ~ 2r-1 / r ~ (-1) UVA UVB(7.2 J/cm2/day 1440 mg / cm ~ (-2) / r ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1)? The skin changes in the back of nude mice were observed with naked eye, and the skin tissues of local lesions in the back of nude mice were cut regularly after 4 weeks, 8 weeks, 12 weeks and 16 weeks of irradiation, and the process of skin changes in the back of nude mice was observed by routine pathological examination and immunohistochemical staining. The expression intensity of CK10 / CK5 / 6 in skin tissue was analyzed. The expression intensity of CK10 / CK5 / 6 in different time points was statistically analyzed by using single factor variance analysis (ANOVA) method with SPSS11.0 software package. [results] there was no obvious abnormal change in the skin of nude mice observed by naked eyes in the blank control group (UVA3. 6 J / cm 2 / 2 day) group. There was no obvious abnormal change in the epithelium of nude mice stained by HE. The skin of nude mice in the UVA UVB(3.6 J/cm2/day 720mJ / cm 2 / day group and UVA UVB(3.6 J/cm2/day 720mJ / cm 2 / day group were proliferated continuously after 16 weeks of irradiation. After 16 weeks of irradiation, the skin on the back of nude mice was obviously proliferated and ruptured. The epithelium showed atypical hyperplasia, a large number of inflammatory cell infiltration or local ulcers (1440mJ / cm ~ 2 / day) and UVA UVB(7.2 J/cm2/day 1440 mg / cm ~ (2) / day). The HE staining for the formation of volcanic or papillary hyperplasia showed that the expression of CK5 / 6CK10 in the UVA irradiation group was similar to that in the normal nude mice skin, and the expression in the UVB or UVA UVB group was increased with the increase of irradiation dose. Its positive expression rate increased gradually. [conclusion] 1 UVB in ultraviolet ray is the main cause of cutaneous squamous cell carcinoma in nude mice. The positive expression rate of CK5 / 6 CK10 was increased with the increase of dose and time, and the expression intensity of CK10 was different in the different time period of ultraviolet induced skin damage and carcinogenesis, with the increase of irradiation dose and irradiation time, the positive expression rate of CK10 was increased gradually.
