湿疹纳米乳膏急性毒性体外替代试验

【摘要】:目的:利用人KC系HaCaT细胞的体外细胞培养,通过四甲基偶氮唑盐比色法(MTT)法测定细胞的增殖活性,得到一组吸光度值(A值),根据吸光度值计算出细胞抑制率IC5。,来检测湿疹纳米乳膏的急性毒性。 方法:采用购自湘雅中心实验室的人KC系HaCaT细胞进行体外细胞传代培养。湿疹纳米乳膏稀释成6个浓度组、及十二烷基硫酸钠的阳性对照组、阴性对照组、空白对照组共9组,分别作用于各组细胞一定时间后用MTT法测定细胞的增殖活性,得到一组吸光度值(OD值),重复实验3次,根据吸光度值计算出细胞抑制率,将细胞抑制率对浓度对数作图,按作图法用SPSS16.0程序计算出IC50。 结果:MTT实验表明:根据吸光度值得出湿疹纳米乳膏6个浓度组的从低到高的细胞抑制率分别为86.017%、85.317%、83.94%、84.14%、87.584%、90.767%,将细胞抑制率对浓度对数作图,按作图法用SPSS16.0计算出:IC50为6.459mg/ml, IC50≥1.5mg/ml,说明湿疹纳米乳膏细胞毒性极小或无毒性。 结论:MTT法检测湿疹纳米乳膏对人KC系HaCaT细胞的细胞增殖的影响,最终实验结果为:湿疹纳米乳膏的IC50值为6.459mg/ml, IC50≥1.5mg/ml,说明湿疹纳米乳膏细胞毒性极小或无毒性,湿疹纳米乳膏经皮吸收和短期应用所产生的毒性反应无或很小。湿疹纳米乳膏应用于临床的急性毒性无或比较小,相对安全,可以用于临床。
[Abstract]:Objective: to determine the proliferation activity of human KC HaCaT cells by MTT assay. A group of absorbance values were obtained and the cell inhibition rate (IC5. 5) was calculated according to the absorbance values to detect the acute toxicity of eczema nanometer cream. Methods: human KC HaCaT cells from Xiangya Central Laboratory were cultured in vitro. Eczema nanometer cream was diluted into 6 concentration groups and 9 groups of positive control group, negative control group and blank control group. After treated with sodium dodecyl sulfate for a certain time, the proliferative activity of the cells was measured by MTT method. A group of absorbance values and OD values were obtained and repeated experiments for 3 times. According to the absorbance values, the cell inhibition rate was calculated. The logarithm of cell inhibition rate against concentration was plotted, and the IC50 was calculated by SPSS16.0 program according to the drawing method. Results: according to the absorbency of eczema nanometer cream, the cell inhibition rates from low to high were 86.01717, 85.31717 and 83.94, respectively.
