尖锐湿疣患者外周血单个核细胞的CXCR3和CCR4表达

【摘要】:目的:测定尖锐湿疣患者外周血趋化因子受体3(CXCR3)和趋化因子受体4(CCR4)在外周血T淋巴细胞的表达,通过其在CD4+T细胞及CD8+T细胞比值的变化,,探讨趋化因子及其受体在尖锐湿疣发病中的作用。方法:采用免疫荧光标记—流式细胞仪检测尖锐湿疣患者和正常人外周血CD4+T细胞及CD8+T细胞的CXCR3和CCR4表达水平,并计算CXCR3和CCR4比值的变化。 结果:尖锐湿疣患者外周血CD4+T淋巴细胞CXCR3的表达与正常对照组比较无显著性差异(P0.05),而CCR4的表达显著增多,具有统计学意义(P0.05);尖锐湿疣患者组CD4+CXCR3/CD4+CCR4比值显著降低(P0.05);尖锐湿疣患者外周血CD8+T淋巴细胞CXCR3的表达水平明显高于正常对照组(P0.01);尖锐湿疣患者外周血CD8+T淋巴细胞CCR4较低,且与正常对照组比较无统计学意义(P0.05),其比值有统计学差异(P0.01);CD4+T淋巴细胞和CD8+T淋巴细胞的CXCR3与CCR4的表达水平均无相关性(P0.05)。 结论:尖锐湿疣患者外周血CD4+T淋巴细胞的CCR4呈高表达,CD8+T淋巴细胞CXCR3表达水平高于正常对照组,表明机体在感染HPV后,细胞免疫状态发生改变,出现尖锐湿疣的患者外周血Th2和CTL细胞增多,一方面可能存在Th1向Th2漂移现象,Th1/Th2失衡,产生免疫逃逸,另一方面,CTL细胞增多有利于机体更好的清除HPV病毒。
[Abstract]:Objective: to determine the expression of chemokine receptor (CXCR3) and chemokine receptor (CCR4) in peripheral blood T lymphocytes of patients with condyloma acuminatum, and to determine the ratio of CXCR3 and CCR4 in CD4 T cells and CD8 T cells. To investigate the role of chemokines and their receptors in the pathogenesis of condyloma acuminatum. Methods: the expressions of CXCR3 and CCR4 in CD4 T cells and CD8 T cells in peripheral blood of patients with condyloma acuminatum and normal subjects were detected by immunofluorescence labeling and flow cytometry, and the ratio of CXCR3 and CCR4 was calculated. Results: there was no significant difference in CXCR3 expression of CD4 T lymphocytes between patients with condyloma acuminatum and normal control group (P 0.05), but the expression of CCR4 was significantly increased in patients with condyloma acuminatum. The ratio of CD4 CXCR3/CD4 CCR4 in patients with condyloma acuminatum was significantly lower than that in patients with condyloma acuminatum, the expression of CXCR3 in peripheral blood CD8 T lymphocytes in patients with condyloma acuminatum was significantly higher than that in normal control group, the CCR4 of CD8 T lymphocytes in patients with condyloma acuminatum was lower than that in patients with condyloma acuminatum. There was no significant difference in the ratio of CXCR3 and CCR4 between P0.01 T lymphocytes and CD8 T lymphocytes compared with the normal control group (P 0.05). Conclusion: the CCR4 expression of CD4 T lymphocytes in patients with condyloma acuminatum is higher than that of normal controls, which indicates that the cellular immune state changes after infection of HPV in patients with condyloma acuminatum. In patients with condyloma acuminatum, the number of Th2 and CTL cells in peripheral blood increased. On the one hand, there may be the imbalance of Th1 / Th2 shift from Th1 to Th2 and immune escape. On the other hand, the increase of CTL cells is beneficial to the elimination of HPV virus.
