FICZ对人皮脂腺细胞芳香烃受体及炎症因子mRNA表达影响及其意义

【摘要】:研究紫外线诱导的光产物FICZ(6-Formylindolo[3,2-b]carbazole)对体外培养的人皮脂腺细胞芳香烃受体(AHR)信号通路影响及相关炎症因子mRNA表达的影响,探讨紫外线诱导痤疮发生的机制。方法:Real-time qPCR检测分析永生化的SZ95人皮脂腺细胞暴露于紫外线光产物FICZ后,检测AHR及其下游基因CYP1A1表达,以及炎症基因IL-1α、IL-1β、RELA等表达。结果:FICZ组AHR表达丰度是CONTROL组(未加入FICZ组,以下均称对照组)的1.412倍(P0.01);FICZ组CYP1A1基因表达丰度是对照组的142.338倍(P0.01);FICZ组IL-1α基因表达丰度是对照组的4.039倍(P0.01);FICZ组IL-1β基因表达丰度是对照组的4.245倍(P0.01);FICZ组RELA基因表达丰度是对照组的1.432倍(P0.01)。结论:紫外线光产物FICZ激活AHR信号通路,从而影响皮脂腺细胞的炎症因子IL-1α、IL-1β、RELA等产生,这可能是紫外线诱导痤疮发生的机制之一。
[Abstract]:To investigate the effect of UV-induced light product FICZ (6-Formylindolo [32-b] carbazole) on the signal pathway of aromatic hydrocarbon receptor (AHR) and the expression of related inflammatory factors in cultured human sebaceous gland cells, and to explore the mechanism of UV-induced acne. Methods the expression of AHR and its downstream gene CYP1A1, and the inflammatory gene IL-1 伪, IL-1 尾 -rela, were detected in SZ95 human sebaceous gland cells exposed to ultraviolet light product (FICZ) by 1: Real-time qPCR. Results the expression abundance of AHR in FICZ group was Contral group (FICZ group was not added to FICZ group). The abundance of CYP1A1 gene expression in FICZ group was 142.338 times higher than that in control group (P0.01). The abundance of IL-1 尾 gene expression in FICZ group was 4.039 times higher than that in control group (P0.01). The abundance of IL-1 尾 gene expression in FICZ group was 4.245 times higher than that in FICZ group (P0.01), and the abundance of rela gene in FICZ group was 1.432 times higher than that in control group (P0.01). Conclusion: ultraviolet light product FICZ activates the AHR signaling pathway, thus affecting the production of IL-1 伪, IL-1 尾 -triarela and so on in sebaceous gland cells, which may be one of the mechanisms of UV induced acne.
