栀子苷及其代谢产物京尼平对人外周血T淋巴细胞的影响

【摘要】:目的:湿疹是常见的炎症性皮肤病,瘙痒剧烈、反复发作,对患者生活影响严重。其发生与Ⅳ型迟发型变态反应有一定关系,T辅助细胞( Th1/ Th2)的失衡在湿疹的发病中起到了重要的作用。前期实验研究和临床应用证明复方茯苓汤治疗湿疹有明显疗效,无明显毒副作用。复方茯苓汤是由茯苓、赤芍、栀子、泽泻、黄柏、当归、浮萍、甘草八味中药组成的复方汤剂,栀子苷是复方茯苓汤的主要活性成分之一,京尼平为栀子苷体内的代谢产物,是栀子苷主要入血成分,两者具有抗炎、解热、利胆和轻泻等多种生物学活性,但对T细胞作用的影响尚不清楚。在炎症反应中,T细胞对免疫识别、免疫应答和免疫调控起着关键的作用。本课题以人外周血T淋巴细胞为模型,探讨复方茯苓汤中有效化学成分栀子苷和京尼平对T细胞增殖和产生Th1或Th2型细胞因子作用的影响,为治疗湿疹及进一步研究开发为治疗新药提供实验依据。 方法:采用SRB法检测PMA活化的人外周血T淋巴细胞的增殖及栀子苷及京尼平对其增殖的抑制,采用ELISA法分析栀子苷和京尼平对活化的人外周血T淋巴细胞上清中IFN-γ和IL-4的影响。 结果:不同浓度栀子苷及京尼平均可以抑制PMA活化的人外周血T淋巴细胞的增殖(P0.01),且呈浓度依赖性。不同浓度的栀子苷及京尼平可显著降低活化人外周血T淋巴细胞上清液中IFN-γ的水平(P0.01),而对活化人外周血T淋巴细胞上清液中IL-4水平的无显著的影响(P0.01)。 结论:复方茯苓汤中栀子苷及其代谢产物京尼平可抑制PMA诱导的人外周血T淋巴细胞的增殖,栀子苷及京尼平可显著降低活化人外周血T淋巴细胞上清液中IFN-γ的水平(P0.01),其抗炎机制可能是通过抑制T淋巴细胞的增殖,降低IFN-γ的水平来实现的。
[Abstract]:Objective: eczema is a common inflammatory skin disease. The pathogenesis of Th _ 1 / Th2 may play an important role in the pathogenesis of eczema. Experimental study and clinical application showed that compound Fuling decoction had obvious curative effect on eczema without obvious side effects. Compound Fuling decoction is a compound decoction composed of Poria cocos, Radix Paeoniae Alba, Gardenia Gardenia, Fructus Alismatis, Cortex Phellodendri, Angelica sinensis, duckweed, Glycyrrhiza and Radix Glycyrrhizae. Geniposide is one of the main active components of compound Fuling decoction, and genipin is the metabolite of Gardenia glycoside. Gardenia glycoside is the main component of blood, both of them have many biological activities, such as anti-inflammatory, antipyretic, choledochal and catharsis, but the effect on T cells is not clear. In inflammatory response, T cells play a key role in immune recognition, immune response and immune regulation. Using human peripheral blood T lymphocytes as a model, the effects of geniposide and geniapine on the proliferation of T cells and the production of Th1 or Th2 cytokines in compound Fuling decoction were studied. For the treatment of eczema and further research and development for the treatment of new drugs to provide experimental basis. Methods: SRB method was used to detect the proliferation of human peripheral blood T lymphocytes activated by PMA and the inhibition of geniposide and geniposide on the proliferation. The effects of geniposide and geniposide on IFN- 纬 and IL-4 in the supernatant of activated human peripheral T lymphocytes were analyzed by Elisa. Results: geniposide and geniposide at different concentrations could inhibit the proliferation of human peripheral blood T lymphocytes activated by PMA (P0.01) in a dose-dependent manner. Different concentrations of geniposide and geniposide significantly decreased the level of IFN- 纬 in the supernatant of activated human peripheral blood T lymphocytes (P0.01), but had no significant effect on the level of IL-4 in the supernatant of activated human peripheral T lymphocytes (P0.01). Conclusion: Geniposide and its metabolite geniposide in compound Fuling decoction can inhibit the proliferation of human peripheral blood T lymphocytes induced by PMA. Geniposide and geniposide significantly decreased the level of IFN- 纬 in the supernatant of activated human peripheral T lymphocytes (P0.01). The anti-inflammatory mechanism may be achieved by inhibiting the proliferation of T lymphocytes and decreasing the level of IFN- 纬.
