日光相关性皮肤肿瘤中DKK1和β-catenin表达的研究

【摘要】:目的: 检测脂溢性角化病、光化性角化病、Bowen病及皮肤鳞状细胞癌患者皮损组织中DKK1和β-catenin的表达情况,探讨DKK1和β-catenin在不同日光相关性皮肤肿瘤中的表达意义及其相关性。 方法: 1.收集在我院皮肤科病理室经临床和病理确诊的脂溢性角化病19例、光化性角化病16例、Bowen病24例及皮肤鳞状细胞癌25例。另外选取22例正常表皮组织标本作为对照,应用免疫组织化学SABC方法检测组织中DKK1和β-catenin的表达。 2.对DKK1和β-catenin的表达进行相关性分析。 结果: 1.DKK1在正常表皮组织的阳性表达定位于胞质中,在SK、AK、BD和SCC中的阳性表达率分别为63.16%、50.00%、12.50%、8.00%。 2.β-catenin在正常表皮组织中的表达定位于胞膜,在SK、AK、BD和SCC中的异常表达率分别为15.79%、56.25%、91.67%、96.00%。 3.皮肤鳞癌中DKK1和β-catenin的表达存在负相关(r=-0.692,P=0.000)。 结论: 1.在表皮恶性转化的过程中,随着病变恶性程度的增加,DKK1的阳性表达率逐渐下降,而β-catenin的异常表达率逐渐上升,提示Wnt信号通路可能在皮肤鳞癌的发生过程中发挥了一定作用。 2.皮肤鳞状细胞癌中DKK1和β-catenin的表达存在负相关,,提示DKK1表达的缺失可能通过作用于经典Wnt信号传导通路促进β-catenin的异常表达。
[Abstract]:Objective: to detect the expression of DKK1 and 尾 -catenin in the lesions of seborrheic keratosis, actinic keratosis and squamous cell carcinoma. To investigate the expression and correlation of DKK1 and 尾-catenin in different sun-associated skin tumors. Methods: 1. Nineteen cases of seborrheic keratosis, 24 cases of photokeratosis and 25 cases of squamous cell carcinoma of skin were collected. The expression of DKK1 and 尾 -catenin in 22 normal epidermal tissues were detected by immunohistochemical SABC method. 2. The correlation between the expression of DKK1 and 尾-catenin was analyzed. Results: the positive expression of 1.DKK1 in normal epidermis was localized in cytoplasm, and the positive expression rate in SKK AKB BD and SCC was 63.1650.The positive expression rate of 尾 -catenin in normal epidermis was 8.000.The expression of 尾 -catenin in normal epidermis was located in the membrane. The abnormal expression rates in SKK AKB BD and SCC were 15.79 and 56.25, 91.67 and 96.00, respectively. There was a negative correlation between the expression of DKK1 and 尾 -catenin in squamous cell carcinoma of skin. Conclusion: 1. In the process of malignant transformation of epidermis, the positive rate of DKK1 decreased gradually with the increase of malignant degree of lesions, while the abnormal expression rate of 尾 -catenin increased gradually. The results suggest that Wnt signaling pathway may play a role in the development of cutaneous squamous cell carcinoma. 2. 2. There is a negative correlation between the expression of DKK1 and 尾 -catenin in squamous cell carcinoma of skin, suggesting that the absence of DKK1 expression may promote the abnormal expression of 尾 -catenin by acting on the classical Wnt signaling pathway.
