木天蓼对VEGF121刺激血管内皮细胞分泌NO的影响

【摘要】:目的探讨木天蓼对血管内皮生长因子121(VEGF121)刺激血管内皮细胞(HUVEC)产生一氧化氮(NO)的影响。方法体外培养HUVEC,分别用VEGF121,VEGF121+木天蓼及实验培养基进行刺激,MTT检测各实验组HUVEC的增殖活力,采用ELISA方法检测培养基中NO的含量。结果 VEGF121能明显刺激HUVEC分泌NO,与对照组相比差异有统计学意义(P=0.044);培养8h后,MTT检测木天蓼+VEGF121组、VEGF121组与对照组相比HUVEC增殖活力差异无统计学意义(P0.05);木天蓼醇提取液(100mg/m L)可抑制VEGF121刺激HUVEC产生NO,差异有统计学意义(P=0.008)。结论VEGF121可促进HUVEC分泌NO,而木天蓼对其具有抑制作用,该结果为木天蓼治疗VEGF相关的皮肤疾病提供了基础理论。
[Abstract]:Objective to investigate the effect of Polygonum hydropiper on the production of nitric oxide (NO) (NO) by vascular endothelial growth factor 121 (VEGF121)-stimulated (HUVEC) in vascular endothelial cells (VEC). Methods HUVECs were cultured in vitro. The proliferative activity of HUVEC in each experimental group was detected with VEGF121VEGF121 and experimental medium, and the content of no in culture medium was detected by ELISA method. Results VEGF121 significantly stimulated HUVEC to secrete no (P0.044), and after 8 hours culture, there was no significant difference in the proliferation activity of HUVEC between VEGF121 group and control group (P0.05). (100mg/m L) could inhibit the production of no in HUVEC stimulated by VEGF121 (P0. 008). Conclusion VEGF121 can promote the secretion of no by HUVEC, but Polygonum hydropiper can inhibit it. The results provide a basic theory for the treatment of VEGF related skin diseases by Polygonum hydropiper.
