尖锐湿疣组织中p-STAT3和COX-2的表达及意义
[Abstract]:Background and purpose
Condylomata acuminatum (CA) is a sexually transmitted disease (STD), caused by human papillomavirus (HPV) infection of genitals, perineum and anus and other parts of a common sexually transmitted disease. Contact transmission. In recent years, the incidence of STD has increased rapidly. According to the reports of STD detection in China, the detection rate of HPV in genitals is 3069/100,000, accounting for the second place of STD in China. In foreign countries, the incidence of HPV is second only to gonorrhea, so it is very important to study its pathogenesis for controlling CA. To explore the pathogenesis of condyloma acuminatum and provide theoretical basis for its prevention and treatment.
Condyloma acuminatum (CA) has the characteristics of rapid proliferation and easy recurrence. Excessive hyperplasia in a few patients has become a giant CA, and some of them can develop tumors. In recent years, studies have found that there are abnormal apoptosis and proliferation in human squamous epithelial cells infected with HPV. There are many studies on its mechanism, but P-STAT3 and COX-2 in CA and genital cancer. In this study, we detected the expression of P-STAT3 and COX-2 in 43 CA tissues and 32 normal prepuce tissues by immunohistochemical SP method, and analyzed the correlation between the expression of P-STAT3 and COX-2 in CA tissues.
Materials and methods
1 material
43 patients with CA were all from the dermatology and venereal disease clinic of the First Affiliated Hospital of Zhengzhou University. The typical lesions were confirmed by histopathology, including 31 males and 12 females, aged 17-55 (31.42 (+ 7.28) years, 20-108 (54.7 (+ 13.63) days, and 32 normal male prepuce excised by urological surgery as control.
2 method
The expression of p-STAT3 and COX-2 in 43 CA tissues and 32 normal prepuce tissues were detected by immunohistochemical SP method. The operation procedure was carried out according to the kit instructions. Grain.
3 statistical processing
SPSS17.0 was used to analyze the expression of P-STAT3 and COX-2 protein in CA tissues. The expression of P-STAT3 and COX-2 protein in CA tissues was compared by rank sum test of two independent samples. The positive expression rate of P-STAT3 and COX-2 protein in CA tissues was compared by x2 test.
Result
Expression of P-STAT3 protein in 1CA and normal epithelial tissue
The positive staining of P-STAT3 was pale yellow to brown granules in the nucleus. 28 of 43 CA tissues were positive for P-STAT3. The positive expression rate was 65.12%(28/43). The expression intensity of P-STAT3 was mostly +++. The P-STAT3 positive cells were mainly located in the nuclei of keratinocytes in the spinous cell layer and granular layer, but not in the normal epithelium. The positive expression intensity of the two groups was significantly different (Z = 5.575, P 0.001).
Expression of COX-2 protein in 2CA and normal epithelial tissue
The positive staining of COX-2 was pale yellow to brown granules in the cytoplasm near the cell membrane and membrane. In 43 CA tissues, 32 cases were positive for COX-2, the positive expression rate was 74.42%(32/43). The expression intensity of COX-2 was mostly +++, and the positive cells were mainly located in the middle and upper part of the spinous layer and granular layer. The positive rate was 6.25% (2/32) and the expression intensity was -++. The difference was statistically significant between the two groups (x2 = 31.71, P 0.05). The difference was statistically significant between the two groups (Z = 5.713, P 0.001).
Correlation analysis of p-STAT3 and COX-2 expression in 3CA tissues
There were 28 positive and 15 negative cases of p-STAT3 expression in 43 CA tissues, 32 positive and 11 negative cases of COX-2 expression. Pearson correlation analysis showed that p-STAT3 expression was positively correlated with COX-2 expression in 43 CA tissues (r=0.577, P 0.001).
conclusion
1. The overexpression of p-STAT3 in CA tissues may be related to the overproliferation of CA cells.
The over expression of COX-2 in 2.CA tissues suggests that COX-2 plays an important role in the development of CA.
3. The expression of p-STAT3 and COX-2 was positively correlated in CA tissues. The co-action of p-STAT3 and COX-2 promoted the occurrence and development of CA.
