β-防御素-2、TNF-a和NF-κB p65在寻常型银屑病皮损中表达的研究

【摘要】：目的：(1)检测β-防御素2 (HBD-2)、TNF-α、NF-кB p65在寻常型银屑病皮损中的表达,研究HBD-2、TNF-α、NF-кB p65的表达与银屑病严重程度的关系；(2)探讨三者在银屑病发生发展中的作用。方法：(1)采用免疫组化SP法(链霉菌抗生物素蛋白-过氧化物酶连接法)检测45例银屑病患者皮损组织、45例银屑病患者非皮损组织及15例正常皮肤组织标本中HBD-2、TNF-α、NF-кB p65的表达,比较不同组别间三者表达的差异,并对HBD-2、TNF-α、NF-кB p65进行相关性分析。(2)根据PASI评分将45例银屑病患者分为轻、中、重三组(轻度25例、中度12例、重度8例),检测轻、中、重银屑病患者HBD-2、TNF-α、NF-кB p65的表达,比较不同组别间三者表达的差异,分析三者在银屑病皮损中的相关性。结果：(1)与正常人皮肤组织和银屑病患者非皮损区相比,银屑病患者皮损区HBD-2、TNF-α、NF-кB p65的表达显著增强,非皮损区HBD-2、TNF-α的表达也高于正常人皮肤组织,差异均有统计学意义(P0.05),非皮损区NF-кB p65的表达与正常人皮肤组织相比差异无统计学意义(P0.05)。在正常皮肤组织、寻常型银屑病非皮损区和银屑病皮损区之间HBD-2与TNF-α, TNF-与NF-кB p65, HBD-2与NF-кB p65均存在正相关(P0.05)。(2)在银屑病皮损区,对HBD-2、TNF-α, NF-кB p65在轻度组、中度组、重度组的表达进行比较,重度组银屑病患者三者的表达高于中度组,中度组又高于轻度组,并具有统计学意义(P0.05)。结论：1、HBD-2, TNF-α在正常皮肤组织、寻常型银屑病非皮损区、寻常型银屑病皮损区中的表达逐渐增强,NF-кB p65在寻常型银屑病皮损区显著高表达。2、HBD-2, TNF-α, NF-кB p65在寻常型银屑病皮损中的表达均升高,且皮损愈重,表达愈强；HBD-2与TNF-α, TNF-α与NF-кB p65, HBD-2与NF-кB p65表达水平之间均存在正相关。综上所述,HBD-2, TNF-α, NF-кB p65与银屑病的发生、发展密切相关,且三者呈协同作用。三者相互促进、相互增强,加重了银屑病皮损。
[Abstract]:Objective: (1) to detect the expression of 尾 -defensin 2 (HBD-2), TNF- 伪 and NF- B p65 in psoriatic lesions, and to study the relationship between the expression of HBD-2,TNF- 伪, NF- B p65 and the severity of psoriasis. (2) to explore the role of three factors in the occurrence and development of psoriasis. Methods: (1) Immunohistochemical SP method was used to detect HBD-2, in 45 cases of psoriasis, 45 cases of psoriasis and 15 cases of normal skin tissue. The expression of TNF- 伪 and NF- B p65 were compared among different groups, and the correlation between HBD-2,TNF- 伪 and NF- B p65 was analyzed. (2) according to PASI score, 45 patients with psoriasis were divided into mild and moderate psoriatic patients. The expression of HBD-2,TNF- 伪 and NF- B p65 in patients with mild, moderate and severe psoriasis were detected in three groups (mild 25 cases, moderate 12 cases, severe 8 cases). To analyze the correlation among the three factors in psoriatic lesions. Results: (1) the expression of HBD-2,TNF- 伪 and NF- B p65 in psoriatic lesions was significantly higher than that in normal skin tissues and psoriatic lesions, and the expression of HBD-2, in non-lesion areas was significantly higher than that in normal skin tissues and psoriatic lesions. The expression of TNF- 伪 was significantly higher than that of normal skin tissue (P0.05), while the expression of NF- B p65 in non-lesion area was not significantly different from that in normal skin tissue (P0.05). In normal skin tissue, there was a positive correlation between HBD-2 and TNF- 伪, TNF- and NF- B p65, HBD-2 and NF- B p65 between psoriasis vulgaris and psoriasis lesions (P0.05). (2). The expression of HBD-2,TNF- 伪 and NF- B p65 in mild group, moderate group and severe group were compared. The expression of HBD-2,TNF- 伪 in severe group was higher than that in moderate group, and the expression in moderate group was higher than that in mild group (P0.05). Conclusion: 1 the expression of HBD-2 and TNF- 伪 in normal skin, psoriasis vulgaris, psoriasis vulgaris and psoriasis vulgaris were gradually increased, and the expression of NF- p65 in psoriasis vulgaris was significantly higher than that in psoriasis vulgaris, and the expression of HBD-2in psoriasis vulgaris was significantly higher than that in psoriasis vulgaris. The expressions of TNF- 伪 and NF- p65 in psoriasis vulgaris were all increased, and the more severe the lesions, the stronger the expression. There was a positive correlation between the expression levels of HBD-2 and TNF- 伪, TNF- 伪 and NF- B p65, HBD-2 and NF- B p65. In conclusion, HBD-2, TNF- 伪, NF- B p65 are closely related to the occurrence and development of psoriasis, and they have synergistic effects. The three promote each other, reinforce each other and aggravate psoriatic lesions.
