皮肤生理指标检测优选特应性皮炎润肤剂的实验研究

【摘要】:目的探讨以神经酰胺、透明质酸钠、丹皮酚、山茶籽油为主要成分研制润肤剂用于治疗特应性皮炎(AD)动物模型,并证实其有效性和安全性。方法按L9(34)正交实验表设计4种主要成分的配比,采用四因素三水平安排实验,将4~6周BALB/c小鼠制备的AD动物模型分为9组,分别涂抹9种主要成分浓度不一的润肤剂,通过检测治疗前及治疗后的皮肤含水量、经皮水分丢失(TEWL)筛选最优处方,并进行药效学及皮肤刺激性实验。结果通过正交表的极差分析以及方差分析得出神经酰胺对皮肤含水量影响最大,山茶籽油对TEWL影响最大,最优的处方为1号,主要成分的配比为D1E1F1G1;药效学实验显示高、中、低剂量组与空白对照组相比治疗后的皮肤含水量、pH值、TEWL值均明显改善,且P值均0.05,与阳性对照组相比无统计学差异,P值均0.05;皮肤刺激性试验结果为0分,无刺激性。结论该润肤剂能明显改善AD小鼠皮肤含水量、pH、TEWL,可用于修复AD小鼠的皮肤屏障,且无刺激性。
[Abstract]:Objective to study the efficacy and safety of ceramide, sodium hyaluronate, paeonol and camellia seed oil in the treatment of atopic dermatitis (AD). Methods according to the L9 (34) orthogonal experiment table, four factors and three levels were used to arrange the experiment. The AD animal model was divided into 9 groups, and 9 kinds of emollients with different concentrations were applied respectively. The optimal prescription was screened by detecting the skin moisture content before and after treatment and skin moisture loss by (TEWL). The pharmacodynamics and skin irritation experiments were also carried out. Results through the range analysis and variance analysis of orthogonal table, it was found that ceramide had the greatest effect on skin water content, and Camellia camellia seed oil had the biggest effect on TEWL, the best prescription was 1, and the proportion of main components was D1E1F1G1. The pharmacodynamics test showed that the skin water content, pH value and TEWL value of the high, middle and low dose groups were significantly improved compared with the blank control group, and the P value was 0.05, and there was no statistical difference between the two groups (P value 0.05). The results of skin irritation test were 0, no irritation. Conclusion the skin moisture content of AD mice can be significantly improved by this moisturizer, and pH,TEWL, can be used to repair the skin barrier of AD mice without irritation.
