不同条件下银屑病患者表皮干细胞分离、培养、鉴定及生物学特性的初步研究

【摘要】:目的:不同条件下,对银屑病患者表皮干细胞(Epidermal Stem Cells,ESCs)体外进行分离、纯化、培养及鉴定,,探索找出最佳培养体系,并对银屑病患者表皮干细胞生物学特性进行初步研究,为银屑病皮损形成的深入研究提供依据。 方法:银屑病皮损标本37例、正常人皮肤标本15例。ESCs分离、筛选、培养:①Dispase酶、胰酶分别4℃过夜或37℃3小时四种消化条件随机选择,进行真表皮分离效率比较;②制备单细胞悬液,比较单位面积银屑病皮损与正常皮肤表皮细胞总数、活细胞数;③IV型胶原不同浓度、不同黏附时间黏附法分选表皮干细胞,计算表皮细胞黏附率;④倒置显微镜下观察细胞生长情况。⑤ESCs表面标志物检测:a.免疫荧光细胞化学法检测原代12h、6d、12d的ESCs角蛋白19(K19)、角蛋白10(K10)的表达;b.流式细胞仪检测初黏附细胞、原代12天、P1代7天ESCs CD29(整合素β1)、CD71、P63的表达。细胞计数法绘制ESCs细胞生长曲线。 结果:ESCs分离、筛选、培养:①2.5g/L Dispase II酶5%CO2、4℃过夜条件下真表皮分离效率高,总高效率达100%。②单位面积的银屑病皮损表皮细胞的数量及活细胞量均大于正常皮肤标本,结果具有显著差异(P<0.05)。③a.IV型胶原黏附表皮细胞10min,可获得纯度较高的ESCs;b.50mg/L与100mg/L浓度IV型胶原表皮细胞黏附率未见显著性差异(P>0.05)。④倒置显微镜下,银屑病ESCs三到四天后开始快速生长,单个银屑病ESCs形成的最大克隆远远大于正常皮肤组,培养数天后,银屑病ESCs易于分化。⑤ESCs表面标志物检测:a.免疫荧光细胞化学法检测:K19显示为亮荧光绿色颗粒沉积在细胞表面及胞浆中,阳性表达;K10未见荧光颗粒沉积;b.流式细胞仪检测CD29、P63高表达,CD71低表达,结合免疫荧光细胞法,认定所培养细胞为表皮干细胞。细胞计数法绘制ESCs生长曲线,结果显示银屑病ESCs短时间内细胞倍增速度快。 结论:①成功分离、筛选、培养银屑病ESCs,条件如下:2.5g/L Dispase II酶4℃过夜消化分离真表皮,2.5g/L胰蛋白酶消化制细胞悬液,50mg/L IV型胶原黏附10min筛选,可成功分选稳定生长的ESCs;②银屑病皮损表皮细胞数量、ESCs增殖能力、细胞生长形态、与正常皮肤存在差异;③银屑病ESCs单细胞克隆能力强,生长迅速,易于分化。
