人脑胶质瘤中ALDH1表达及其与Ki-67和MGMT的关系研究

【摘要】:目的研究ALDH1、Ki-67和MGMT在人脑胶质瘤组织中的表达与胶质瘤病人的临床资料的关系;探讨ALDH1、Ki-67和MGMT三者之间的关系,并阐明它们与胶质瘤恶性程度的关系;联合检测ALDH1、Ki-67和MGMT蛋白,为准确地诊断胶质瘤的恶性程度提供一种方法,同时,也为胶质瘤的个体化治疗提供理论依据。 方法连续收集兰州大学第二附属医院神经外科临床中心2012年7月至2013年10月间经手术治疗的胶质瘤病人的病理标本及临床资料53例,纳入研究,所有病例均为新发诊断为胶质瘤的病人,排除复发的病例、以及之前施行化疗、放疗等治疗的病例。采用免疫组织化学的方法检测胶质瘤标本中ALDH1、Ki-67和MGMT蛋白的表达情况,采用SPSS18.0统计学软件分析ALDH1、Ki-67和MGMT蛋白在人脑胶质瘤组织中的表达及意义。 结果本研究显示,患者的性别、年龄及肿瘤组织学类型与胶质瘤组织中ALDH1、Ki-67和MGMT的表达无关。高级别组(WHOⅢ-Ⅳ级)中ALDH1、Ki-67和MGMT的表达均较低级别组(WHO Ⅰ-Ⅱ级)显著升高(ALDH1,93.10%比70.83%,p0.01;Ki-67,96.55%比70.83%,p0.01;MGMT,72.41%比50.00%,p0.01)。另外,本研究发现人脑胶质瘤中ALDH1、Ki-67和MGMT三者之间呈高度显著正相关关系(ALDH1与Ki-67, rs=O.904, p=0.000; ALDH1与MGMT, rs=0.833, p=0.000; Ki-67与MGMT, rs=0.779, p=0.000)。 结论结果表明,在人脑胶质瘤中,ALDH1,Ki-67和MGMT蛋白的表达与胶质瘤的病理级别相关,级别越高,三种蛋白的表达就越强。ALDH1、Ki-67和MGMT三者之间高度显著正相关关系,表明胶质瘤中ALDH1、Ki-67和MGMT蛋白三者之间的内在一致性联系,联合检测三个指标可能更能准确地判断胶质瘤患者恶性程度和预后,对胶质瘤患者制定个体化的治疗方案可能有着重要的参考价值。
[Abstract]:Objective to study the relationship between the expression of ALDH1 Ki-67 and MGMT in human glioma tissues and the clinical data of glioma patients. To explore the relationship between Ki-67 and MGMT in ALDH1, and to clarify their relationship with the malignant degree of glioma. The combined detection of ALDH1 Ki-67 and MGMT protein provides a method for the accurate diagnosis of malignant degree of glioma and also provides theoretical basis for individualized treatment of glioma. Methods the pathological specimens and clinical data of 53 patients with glioma treated surgically from July 2012 to October 2013 in the Neurosurgery Clinical Center of the second affiliated Hospital of Lanzhou University were collected. Included in the study, all cases were newly diagnosed as gliomas, excluding recurrent cases, and prior chemotherapy. Immunohistochemical method was used to detect the expression of ALDH1Ki-67 and MGMT protein in gliomas. The expression and significance of ALDH1 Ki-67 and MGMT protein in human gliomas were analyzed by SPSS18.0 software. Results in this study, sex, age, tumor histological type and ALDH1 in glioma tissues were found.
