卒中后应激对大鼠认知功能与海马MAP2表达水平的影响

【摘要】:目的:①研究卒中后应激对大鼠认知功能的影响。②明确卒中后应激对大鼠海马微管相关蛋白2(microtubule associated protein2,MAP2)表达水平的影响。 方法:①采用大脑中动脉栓塞(middle cerebral artery occlusion,MCAO)术制备卒中模型。②抑郁症处理采用卒中后给予慢性不可预期性温和应激(chronic unpredictable mild stress,CUMS)连续处理3周。③Y迷宫(Y-maze)、新颖物体识别(novel objective recognition,NOR)实验评估卒中后抑郁症大鼠的认知功能。④蛋白免疫印迹(western blot,WB)观察海马MAP2的表达。 结果:①与sham组(即假手术组)相比较,缺血或者慢性应激可使大鼠体重、蔗糖偏好量、旷场实验水平运动及旷场实验垂直运动明显下降,二者的综合作用更明显。②缺血或者慢性应激可使大鼠认知功能损伤,表现为与同时间点的sham组(即假手术组)相比较,大鼠Y迷宫试验的正确交替率下降和新颖物体识别试验中辨别指数亦下降,二者的综合作用更明显。③缺血或者慢性应激可使大鼠海马MAP2的表达下调,表现为与sham组(即假手术组)相比较,MCAO组(即单纯卒中组)和stress组(即单纯抑郁症组)大鼠海马MAP2的表达下调,二者的综合作用PSD组(即卒中后抑郁症组)大鼠海马MAP2的表达下调更明显。 结论:①卒中后应激加重卒中大鼠认知功能损伤;②卒中后应激下调大鼠海马区MAP2的表达水平。
[Abstract]:Objective to investigate the effects of post-stroke stress on cognitive function in rats. 2 to investigate the effect of post-stroke stress on the expression of microtubule-associated protein 2 associated protein 2 (MAP2) in rat hippocampus. Methods the middle cerebral artery occlusion of middle cerebral artery (MCAO) was used to make stroke model .2 depression was treated with chronic unpredictable mild stress (chronic unpredictable mild stress CUMS2) for 3 weeks. 3 Y maze to recognize novel objective. The cognitive function of poststroke depression rats was evaluated by recognition nor. The expression of hippocampal MAP2 was observed by Western blotWBs. Results compared with sham group (sham operation group), ischemia or chronic stress significantly decreased body weight, sucrose preference, horizontal exercise in open field experiment and vertical movement in open field experiment. The combined effects of the two groups were more obvious. 2. 2 Ischemia or chronic stress could induce cognitive impairment in rats, which was compared with the sham group at the same time point (that is, the sham operation group). The correct alternation rate of Y maze test decreased and the discriminant index in novel object recognition test decreased. The combined effect of these two factors was more obvious. 3. 3 ischemia or chronic stress could down-regulate the expression of MAP2 in hippocampus of rats. Compared with sham group (sham operation group), the expression of MAP2 in hippocampus was down-regulated in MCAO group (simple stroke group) and stress group (simple depression group). The down-regulation of MAP2 expression in hippocampus of PSD group (post-stroke depression group) was more obvious. Conclusion stress after stroke exacerbates cognitive impairment in stroke rats. (2) Post-stroke stress down-regulates the expression of MAP2 in hippocampal area of rats.
