外源性神经球蛋白与氯化血红素对大鼠局灶性缺血脑组织的联合保护作用

【摘要】:目的探讨外源性神经球蛋白与氯化血红素对大鼠缺血脑组织的联合保护作用及其可能机制。 方法将175只SD大鼠随机分为5组(n=35),分别为假手术组,缺血组,氯化血红素干预组,质粒干预组,氯化血红素及质粒共同干预组。采用大鼠MCAO脑缺血模型,麻醉清醒后,行神经行为学检测,术毕24h检测脑组织含水量、脑梗死体积比、原位细胞凋亡技术检测神经元凋亡、免疫荧光技术检测Bcl-2蛋白表达及Western蛋白印迹技术分析Ngb蛋白、Bcl-2蛋白表达情况。 结果质粒携带的外源性Ngb与Hemin联合干预组在神经行为学评分、脑组织含水量、脑梗死体积比方面显著降低,凋亡神经元数量减少,同时皮层神经元Ngb及Bcl-2蛋白表达增加,均较单独干预组有显著差异(p0.05)。 结论在大鼠局灶性脑缺血条件下,外源性Ngb与Hemin联合作用,可能通过增强Ngb蛋白的表达,实现Bcl-2蛋白表达增加,抑制皮层神经元凋亡,显著降低了凋亡神经元数目、脑组织梗死体积比、脑组织含水量和神经行为评分,,从而获得对局灶性缺血后神经元的更佳保护作用。
[Abstract]:Objective to investigate the combined protective effect of exogenous neuroglobulin and hemin on ischemic brain tissue in rats and its possible mechanism. Methods 175 Sprague-Dawley rats were randomly divided into 5 groups: sham operation group, ischemia group, hemin intervention group, plasmid intervention group, hemin and plasmid co-intervention group. The brain water content, the volume ratio of cerebral infarction and the apoptosis of neurons were detected by in situ apoptosis technique at 24 hours after operation. The expression of Bcl-2 protein was detected by immunofluorescence technique and the expression of Bcl-2 protein by Ngb protein was analyzed by Western Western blot. Results the neurobehavioral score, brain water content, cerebral infarction volume ratio, the number of apoptotic neurons and the expression of Ngb and Bcl-2 protein in cortical neurons were significantly decreased in the intervention group with exogenous Ngb and Hemin. There was significant difference between the two groups (P 0.05). Conclusion under the condition of focal cerebral ischemia, the combination of exogenous Ngb and Hemin may enhance the expression of Ngb protein, increase the expression of Bcl-2 protein, inhibit the apoptosis of cortical neurons, and decrease the number of apoptotic neurons. The cerebral infarction volume ratio, brain water content and neurobehavioral score were used to obtain a better protective effect on neurons after focal ischemia.
